摘要
通过对我国羊巴贝斯虫bov57基因进行克隆、序列比对和进化分析,为研究bov57基因功能及我国羊巴贝斯虫分类提供基础数据支持。以NCBI数据库中公布的卵形巴贝斯虫和牛巴贝斯虫bov57基因序列BLAST我国2个羊巴贝斯虫基因组数据库,获得羊巴贝斯虫bov57基因的保守序列,以此为模板设计引物;扩增我国6株羊巴贝斯虫的bov57基因,分析其结构特征,并进行序列比对,构建系统进化树,分析遗传进化关系。结果显示,这6株羊巴贝斯虫bov57基因均无内含子,羊巴贝斯虫未定种敦煌株/新疆株(BspDH/XJ)和莫氏巴贝斯虫临潭株/天祝株(Bm LT/TZ)bov57基因ORF的大小为1608 bp,编码536个氨基酸;莫氏巴贝斯虫河北株/宁县株(Bm HB/NX)bov57基因ORF的大小为1605 bp,编码535个氨基酸。序列比对结果显示,羊巴贝斯虫未定种与莫氏巴贝斯虫bov57基因核苷酸序列的相似性为58.84%~61.71%,氨基酸序列的相似性为45.93%~50.93%;莫氏巴贝斯虫临潭株/天祝株和莫氏巴贝斯虫河北株/宁县株的核苷酸序列相似性为85.29%~85.60%,氨基酸序列相似性为75.19%~75.74%。遗传进化分析结果表明,我国的这6株羊巴贝斯虫分为2个种,而莫氏巴贝斯虫可能存在2个亚种。
Through performing the gene cloning,sequence alignment and phylogenetic analysis of ovine Babesia bov57 gene,it provides the support of foundational data for researching bov57 gene function and classification of ovine Babesia species in China.On the basis of bov57 gene sequences of Babesia bovis and Babesia ovata,the conserved sequences were obtained by BLAST to local genomic databases of Babesia sp.Xinjiang and Babesia motasi Lintan strains.Primer pairs were generated using these conserved sequences as templates.Full-length bov57 gene sequences were amplified from six Chinese ovine Babesia strains,respectively,and their gene structural feature and evolutionary relationship were analysed.These results showed that no intron was observed in bov57 genes of six Chinese ovine Babesia.The sizes of bov57 gene ORFs were 1608 bp,encoding 536 amino acids,in Babesia sp.Dunhuang/Xinjiang(Bsp DH/XJ)and B.motasi Lintan/Tianzhu(Bm LT/TZ)and 1605 bp,encoding 535 amino acids,in B.motasi Hebei/Ningxian(Bm HB/NX).Sequence alignments showed that nucleotide sequence similarities of B.motasi and Babesia sp.strains were ranged from 58.84%—61.71%,while those of the amino acid sequences were45.93%—50.93%.The nucleotide sequence similarities among B.motasi Bm LT/TZ and Bm HB/NX were 85.29%—85.60%and those of amino acids were 75.19%—75.74%.The results of phylogenetic analysis showed that the six Chinese ovine Babesia strains were divided into two species and it’s possible that there were two sub-species in B.motasi.
