摘要
目的观察参芎葡萄糖注射液对大鼠肺动脉压、肺血管重构、右心室肥厚及肺组织脂联素(adiponectin,APN)蛋白表达的影响,探讨参芎葡萄糖注射液对大鼠肺动脉高压(pulmonary hypertension,PH)的作用及其机制,为其治疗PH提供实验依据。方法采用野百合碱(monocrotaline,MCT)按60 mg/kg剂量单次腹腔注射建立PH SD大鼠模型,将模型大鼠分为MCT组和参芎葡萄糖注射液治疗组,参芎葡萄糖注射液治疗组腹腔注射单剂量MCT后第2~28天,每日按19.5 g/kg剂量注射参芎葡萄糖注射液1次。另取20只大鼠作为正常对照组,按60 mg/kg经腹腔单剂量注射生理盐水。各组大鼠饲养4周(28 d)后,检测平均肺动脉压(mean pulmonary artery pressure,mPAP)和右心肥厚指数(right ventricular hypertrophy index,RVHI),光镜下观察肺血管结构改变,免疫组化及Western blot法检测肺组织APN蛋白的表达。结果MCT组较正常对照组大鼠肺小动脉平滑肌明显增生肥大,而参芎葡萄糖注射液治疗组大鼠肺小动脉平滑肌增生肥大较MCT组有所改善;参芎葡萄糖注射液治疗组大鼠较MCT组m PAP明显降低(P<0.01),RVHI明显下调(P<0.01),APN蛋白表达明显上调(P<0.01)。结论参芎葡萄糖注射液能改善PH大鼠的mPAP及右心肥厚,并上调其肺组织APN的表达,提示其发挥对PH的保护作用过程中可能有APN蛋白的参与。
Objective To observe the influence of Shenxiong Glucose Injection on pulmonary artery pressure,pulmonary vascular remodeling,right ventricular hypertrophy and expression of adiponection(APN)in pulmonary tissue of rats and investigate the effect of Shenxiong Glucose Injection on pulmonary hypertension(PH)as well as the relevant mechanism so as to provide an experimental basis for the treatment of PH.Methods SD rat model of PH was established by a single intraperitoneal injection with monocrotaline(MCT)at a dosage of 60 mg/kg.The model rats were divided into MCT control and treatment groups.The rats in treatment group were injected i.p.with Shenxiong Glucose Injection at a dosage of 19.5 g/kg once a day on days 2~28 after injection with MCT,while those in MCT control group were untreated.Another 20 rats were served as normal control and injected with a single dose of 60 mg/kg physiological saline.The rats in various groups were raised for 4 weeks(28 d),of which the mean pulmonary artery pressure(mPAP)and right ventricular hypertrophy index(RVHI)were measured,the structural changes of pulmonary arteries were observed by light microscopy,and the expression of APN protein in pulmonary tissue was determined by immunohistochemical assay and Western blot.Results Obvious hyperplasia and hypertrophy of smooth muscle of pulmonary arteriole were observed in MCT control group as compared with those in normal control group,while were improved significantly in treatment group.However,as compared with that in MCT control group,the mPAP in treatment group decreased significantly,the RVHI was significantly down-regulated,and the expression of APN protein was significantly up-regulated(each P<0.01).Conclusion Shenxiong Glucose Injection improved the mPAP and right ventricular hypertrophy of rats with PH and upregulated the expression of APN protein,indicating that APN protein might involve in the protection against PH.
