摘要
为了探究广西南宁市冬季规模化鸡场环境中细菌种类及分布情况,采集了鸡场饮水、饲料和空气样品,进行细菌的分离培养。根据菌落生长的不同形态特征,进行菌株纯化,对纯化菌株的16S rDNA基因进行PCR扩增并测序分析。结果显示,从不同样品中共分离纯化细菌菌株71株,小鸡舍内小鸡饲料、空气环境中的细菌及饮水中的细菌总数分别是7.0×10^4 CFU/g、3.3×10^3~1.9×10^4 CFU/m3和1.04×10^2 CFU/mL,均未超过国家标准;将测序获得的16S rDNA基因序列在NCBI上进行比对后,确定鸡舍环境中含有大肠埃希菌、金黄色葡萄球菌等20种条件性致病菌,木糖葡萄球菌、地衣芽胞杆菌等8种非致病菌。试验结果为改善鸡舍内环境的空气质量和细菌性疾病的预防提供数据参考。
To explore the species and distribution of bacteria from the environment of large-scale chicken farm in winter in Nanning,Guangxi province,the samples from drinking water,feed and air of chicken farm were collected,and the bacteria of samples were isolated and cultured.The strains were purified according to the different morphological characteristics of the colony growth,and the 16S rDNA genes of the purified strains were amplified by PCR and sequenced.The results showed that 71 strains of bacteria were isolated and purified.The total number of bacteria in chicken feed,air environment and drinking water of chicken house were 7.0×10^4 CFU/g,3.3×10^3-1.9×10^4 CFU/m3 and 1.04×10^2 CFU/mL,respectively,which did not exceed the national standard.Comparison results of the sequences of 16S rDNA genes in NCBI showed that there were 20 kinds of conditional pathogenic bacteria including Escherichia coli and Staphylococcus aureus,and so on.8 kinds of non-pathogenic bacteria such as Staphylococcus xylosus and Bacillus licheniformis were found in the environment of chicken house.The results of this study provided a reference for improving the air quality and the prevention of bacterial diseases in chicken houses.
