摘要
动情周期和应激会影响雌性动物对成瘾药物的敏感性。糖皮质激素受体(GR)和精氨酸加压素(AVP)参与调节应激反应和药物成瘾。论文探讨了慢性束缚应激是否影响雌性小鼠动情周期继而影响可卡因诱导的行为及GR、AVP和皮质酮(CORT)的反应。结果显示,束缚10 d(2 h/d)的小鼠动情期延长,束缚24 d(2 h/d)的小鼠动情前期、动情期及动情后期延长,且动情间期缩短。束缚24 d后,连续注射可卡因(2次/d,20 mg/kg/次)2 d后检测发现,与盐水对照相比,可卡因增加了束缚鼠和非束缚鼠的运动性,但慢性束缚没有改变可卡因诱导的运动性及焦虑水平。慢性束缚虽增加了盐水组血清中的CORT浓度,并降低了海马GR-ir神经元及下丘脑室旁核(PVN)和视上核(SON)的AVP-ir神经元表达,但对可卡因组没有显著影响。结果表明,慢性束缚应激虽然改变了雌性动情周期及相关的神经内分泌反应,但没有改变可卡因诱导的运动性及GR、AVP和CORT的反应。
The estrous cycle and stress contribute to the action of psychostimulant drugs in females.Glucocorticoid receptors(GR)and arginine vasopressin(AVP)are the main mediators of stress response and drug abuse.In the present study,we examined whether chronic restraint stress(CRS)affects estrous cycle and cocaine-induced behavioral and GR,AVP and corticosterone(CORT)responses in female mice.Female mice were subjected to restraint stress 2 h daily for 10 days or 24 days.The results showed that the estrus duration was longer in the 10 d CRS group compared with the control group.The duration of the proestrus,the estrus and the metestrus were prolonged,while the duration of the diestrus was shortened in the 24 d CRS group.Following continuous cocaine injection for 2 days(2 times/d,20 mg/kg/time,i.p.)after 24 d CRS,we found cocaine increased locomotion in restraint and nonrestraint groups.CRS exerted no effects on cocaine-induced locomotion and anxiety levels.CRS up-regulated levels of serum CORT and reduced GR-ir expression in the hippocampus and AVP-ir expression in the paraventricular nucleus(PVN)and supraoptic nucleus(SON)in saline control,but did not change these measures in cocaine-treated group.These results indicate that CRS alters estrous cycle and associated with neuroendocrine responses,although does not affect cocaine-induced locomotion and the responses of the GR,CORT and AVP.
