猪源伪狂犬病病毒传代致弱LA2017株的安全性和免疫效力研究 被引量:4

SAFETY AND IMMUNOGENICITY OF THE ATTENUATED VIRUS LA2017 DERIVED FROM PSEUDORABIES VIRUS EPIDEMIC STRAIN

摘要 为检测猪源伪狂犬病病毒变异株AH02LA株作为疫苗后选株的安全性及免疫效力。本研究对4日龄初生仔猪和产前1个月妊娠母猪免疫LA2017株后的安全性、28~35日龄仔猪免疫效力以及免疫猪后排毒和同群传播性进行了研究。结果显示:滴鼻接种4日龄仔猪和肌肉注射产前1个月的妊娠母猪,均无任何临床症状,表明该疫苗株具有良好的安全性;肌肉注射接种28~35日龄的仔猪,免疫后7 d产生对PRV变异株AH02LA的完全保护并且可以阻止排毒,免疫14 d后攻毒均未发病和排毒,而PRV Bartha K61株免疫猪后7 d攻毒,有4头猪出现了体温升高,鼻拭子样品均检出病毒,免疫后14 d和21 d时对试验猪攻毒,PRV Bartha K61株免疫组均有猪发病,且鼻拭子样品均检出病毒;在抗体产生水平方面,PRV LA2017株免疫后14 d产生高水平中和抗体,PRV中和指数抗体≥10 000,且维持至免疫后5个月,而Batha-K61组免疫后1个月至3个月针对PRV变异株AH02LA的中和指数仅为178~1000,且4个月就开始下降;在同群感染方面,PRV LA2017株肌肉注射接种28~35日龄仔猪后14 d内均未从鼻拭子和肛门拭子检出排毒,gB抗体全部转为阳性,同群非免疫接种猪PRV gB和gE的ELISA抗体均为阴性,表明PRV LA2017株安全性好,无横向传播,不引起同群感染。结果表明,PRV LA2017株作为疫苗株对猪伪狂犬变异株的保护效果显著优于Bartha K61株,该毒株安全性好、抗体水平高和持续时间长,是一株极具开发价值的猪伪狂犬病流行株的弱毒疫苗候选株。 The PRV LA2017 strain was attenuated by serial passages of the Pseudorabies virus mutant strain AH02LA on CEF cells with the loss of the gE gene.This study examined the safety of the attenuated virus strain PRV LA2017 in 4-day-old newborn piglets and pregnant sows of 1-month before parturition and immunogenicity in 28-35 days-old piglets.The results showed that intranasal immunization for 4-day-old newborn piglets and pregnant sows with titers of 107,106,and 105 TCID50 of the attenuated strain did not cause any clinical signs,indicating that the attenuated strain PRV LA2017 was safe.The 28-35 days-old piglets was inoculated with the PRV LA2017 strain and all piglets were protected and no viruses were shed after challenge with PRV mutant strain AH02LA.Meanwhile,28-35 days-old piglets were inoculated with the PRV Bartha K61 strain and subsequently challenged with PRV mutant strain AH02LA at days 7 post immunization.Four out of these piglets showed high body temperature and virus shedding by detection of their nasal swabs although they had no clinical signs.When these piglets were challenged at 14 and 21 days post immunization,all piglets got sick and virus shedding.In addition,piglets immunized with PRV LA2017 strain produced high level neutralizing antibodies at day 14 post immunization,and the antibody neutralization index of PRV reached 10 000 or higher,which lasted for 5 months.In contrast,the neutralization index of PRV in the Bartha K61 vaccine immunized piglets was 178-1000 at 1 to 3 months and all piglets had diseases within 4 months post immunization.Meanwhile,gB antibody was positive and no viremia was detected from nasal and anal swabs in the piglets immunized with the PRV LA2017 strain.However,the gB and gE antibodies were negative in the sentinel piglets that were not inoculated with the PRV LA2017 strain.These results indicated that intramuscular injection the PRV LA2017 strain did not shed virus causing transmission to non-immunized piglets.In conclusion,the PRV LA2017 strain showed a significantly b
