摘要
目的:检测瘢痕疙瘩中微小RNA-34b(miR-34b)的表达,分析其与转化生长因子-β1(TGF-β1)的相关性。方法:收集42例瘢痕疙瘩术后组织作为观察组,收集42例增生性瘢痕术后组织作为对照组,收集正常皮肤组织42例作为正常对照组。应用实时荧光定量PCR法检测三组中miR-34b的表达,应用免疫组化和Western Blot法检测观察组中TGF-β1的表达。结果:观察组中miR-34b的表达明显低于对照组和正常对照组,差异有统计学意义(P<0.05)。观察组中miR-34b的表达在有无家族史及不同病变高度的分组中差异有统计学意义(P<0.05);而在不同性别、年龄、病程的分组中差异无统计学意义(P>0.05)。相关分析显示miR-34b与TGF-β1呈负相关性。结论:瘢痕疙瘩中miR-34b低表达,与家族史及病变增生的高度相关。miR-34b与TGF-β1具有协同负向作用。
Objective To detect the expression of microRNA-34b(miR-34b)in keloid,analyze its correlation with transforming growth factor-β1(TGF-β1).Methods 42 cases of keloid were collected as the observation group.42 cases of hypertrophic scar were collected as the control group.42 cases of normal skin were collected as the normal control group.The expression of miR-34b was detected by real-time fluorescent quantitative PCR,and TGF-β1 was detected by immunohistochemistry and Western Blot method.Results Expression of miR-34b was significantly lower in the observation group than that in the control group and the normal control group,the difference is statistically significant(P<0.05).The expression of miR-34b was statistically significant in family history and different lesion hight(P<0.05),but there was no significant difference in gender,age and course of disease(P>0.05).Correlation analysis showed that miR-34b was negatively correlated with TGF-β1.Conclusion Lower expression of miR-34b is related to family history and lesion height.MiR-34b and TGF-β1 have synergistic negative effects.
