miR-4472的靶基因预测及生物信息学分析

摘要 目的应用生物信息学分析方法预测miR-4472的靶基因及信号通路。方法利用miRBase数据库分析miR-4472的物种保守性;利用Targetscan、miRDB数据库进行靶基因预测,再用DAVID数据库将miR-4472的预测靶基因集合进行基因本体(GO)功能富集分析及京都基因与基因组百科全书(KEGG)生物通路富集分析。结果miR-4472预测的交集靶基因共378个。GO分析结果显示,miR-4472的靶基因主要参与转录的正负调节、细胞凋亡、蛋白质磷酸化等生物过程(均P<0.01)。KEGG信号通路分析结果显示,miR-4472的靶基因显著富集于PI3K-Akt信号通路、胰高血糖素相关信号通路、心肌细胞中的肾上腺素能信号传导通路、醛固酮的合成和分泌相关信号通路(均P<0.05)。结论miR-4472靶基因富集的信号通路均与新生儿缺氧缺血性脑病密切相关,尤其是P13K/AKt信号通路。 Objective To predict the target genes and signaling pathways of miR-4472 through bioinformatics analysis.Methods MiRBase was used to analyze the species conservation of miR-4472.And the target genes of miR-4472 were predicted with Targetscan and miRDB.Then the function of target genes were analyzed by gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)pathway analysis through the DAVID database.Results A total of 378 target genes of miR-4472 were predicted.Go analysis showed that the target genes of miR-4472 were mainly involved in biological processes such as positive and negative regulation of transcription,apoptosis,and protein phosphorylation(P<0.01).The KEGG pathway analysis showed that the target genes of miR-4472 were significantly enriched in PI3K-Akt signaling pathway,glucagon signaling pathway,adrenergic signaling in cardiomyocytes and aldosterone synthesis and secretion pathways(P<0.05).Conclusion The signaling pathways for miR-4472 target genes enrichment are closely related to hypoxic-ischemic encephalopathy in newborns,especially the P13K/AKt signaling pathway.
