摘要 目的探讨过氧化物氧化还原酶蛋白1(PRDX1)在乳腺癌组织中的表达及对乳腺癌细胞生长调控的机制。方法选择2017年4月至2019年2月在北大医疗鲁中医院进行手术治疗的86例乳腺癌患者为研究对象,分析PRDX1表达与乳腺癌患者临床病理特征的关系。培养MCF7乳腺癌细胞株,分组转染成空白对照组、siRNA-NC组、PRDX1-siRNA转染组。检测不同组细胞增殖抑制率及凋亡率,采用Western blot检测PI3K/AKT信号通路相关蛋白表达水平。结果①PRDX1在乳腺癌及癌旁组织中的阳性率为58.1%(50/86)、23.3%(20/86)、在正常组织中的阳性率为3.7%(3/82),PRDX1在乳腺癌癌组织中的阳性率较高,且PRDX1阳性表达与乳腺癌患者的TNM分期、病理分级有关(P<0.05)。②TT检测结果显示,在24 h、48 h、72 h时,siRNA-NC组的细胞抑制率分别为1.6%、1.7%、2.4%,PRDX1-siRNA组分别为5.6%、15.3%、38.4%,PRDX1-siRNA组的细胞抑制率高于空白组与siRNA-NC组(P<0.05)。③流式细胞检测显示,空白组、siRNA-NC组、PRDX1-siRNA组的凋亡率分别为(7.7±1.3)%、(7.4±1.5)%、(25.7±4.2)%,PRDX1-siRNA的肿瘤凋亡率最高。④Western blot检测显示,与空白组相比,siRNA-NC组的PI3K蛋白、AKT蛋白表达量无明显变化,但PRDX1-siRNA组中PI3K蛋白、AKT蛋白的表达量下降(P<0.05)。结论PRDX1在乳腺癌患者中的阳性率明显提高。在MCF7乳腺癌细胞株中,抑制PRDX1表达水平可通过降低PI3K/AKT信号通路活性,促进乳腺癌细胞凋亡。 Objective To investigate the expression of PRDX1 in breast cancer and its mechanism of regulating the growth of breast cancer cells.Methods Eighty-six patients with breast cancer who underwent surgery in PKUCare Luzhong Hospital from April 2017 to February 2019 were selected.The relation between the expression of PRDX1 and clinicopathologic features of breast cancer patients was analyzed.MCF7 breast cancer cell lines were cultured and divided into blank control group,siRNA-NC group and PRDX1-siRNA transfection group.Cell proliferation inhibition rate and apoptotic rate were detected.And the expression level of PI3K/AKT signaling pathway related protein was detected by Western blot.Results The positive rates of PRDX1 in breast cancer and adjacent tissues were 58.1%and 23.3%,which was 3.7%in normal tissues.The positive rate of PRDX1 in breast cancer patients was related to the TNM staging and pathological grading.At 24 h,48 h and 72 h,the cell inhibition rates in siRNA-NC group were 1.6%,1.7%and 2.4%,which were lower than those of RDX1-siRNA group(5.6%,15.3%and 38.4%)significantly(P<0.05).The apoptotic rates of blank control group,siRNA-NC group and PRDX1-siRNA transfection group were(7.7±1.3)%,(7.4±1.5)%,(25.7±4.2)%.The apoptotic rate of RDX1-siRNA was the highest.Compared with the blank group,there was no significant change in the expression of PI3K and AKT protein in siRNA-NC group.However,the expression of PI3K and AKT protein in RDX1-siRNA group decreased significantly(P<0.05).Conclusions Inhibition of PRDX1 expression in MCF7 breast cancer cell lines can promote apoptosis of breast cancer cells by reducing the activity of PI3K/AKT signaling pathway.
