摘要
葵花籽蛋白富含芳香族氨基酸。本研究旨在考察其作为食物主要蛋白来源时,对抑郁模型小鼠大脑多巴胺(dopamine,DA)和五羟色胺(5-hydroxytryptamine,5-HT)产生的影响,及缓解抑郁发生的作用。将C57BL/6J小鼠分别用大豆蛋白(模型对照,MOD)、葵花籽蛋白(SFSP)和乳清蛋白组(WPC,阳性对照)为主要蛋白源饲料饲养2月,进行慢性不可预知温和应激(chronic unpredictable mild stress,CUMS)造模处理,用未造模动物作为正常对照(CON,10只/组)。SFSP显著预防CUMS引起的糖水偏好率降低、悬尾或强迫游泳不动时间延长,预防在旷场和高架十字迷宫的焦虑行为,效果与WPC相当。SFSP也提高了大脑海马组织5-HT、DA、去甲肾上腺素水平和脑源性神经营养因子,提高了血清色氨酸、苯丙氨酸、酪氨酸浓度和色氨酸与中性氨基酸比值。同时降低了血清皮质酮水平,提高抑郁小鼠皮层中超氧化物歧化酶、谷胱甘肽过氧化物酶和过氧化氢酶活性,降低丙二醛和活性氧自由基水平。SFSP通过增加芳香族氨基酸吸收、提高海马单胺类神经递质浓度,和预防大脑皮层氧化应激发挥预防抑郁发生的作用。
Sunflower seed protein(SFSP) is rich in aromatic amino acids.The purpose of this study was to explore the effect on production of dopamine(DA) and 5-hydroxytryptamine(5-HT) in brain and the antidepressant-like effect of SFSP when it was used as the main protein source of dietary food.C57 BL/6 J mice were fed with food which soy protein(model control,MOD),sunflower seed protein(SFSP),and whey protein(WPC,positive control) as the main protein source,respectively,for 2 months.The chronic unpredictable mild stress(CUMS) model was established to induce depression in mice.The normal group(CON,10 animals/group) was not exposed to the CUMS model.SFSP significantly prevented the decrease of sucrose preference rate,the increase of immobility time in the tail suspension test and forced swimming test and improved anxiety behavior in the open field test and elevated plus maze test caused by CUMS.The effect was not different from whey protein.SFSP also increased the levels of 5-HT,DA,norepinephrine and brain-derived neurotrophic factor in hippocampus,increased serum tryptophan,phenylalanine,tyrosine concentrations and the ratio of tryptophan over large neutral amino acids.At the same time,it reduced the level of corticosterone in plasma,increased the superoxide dismutase,glutathione peroxidase and catalase activity,and reduced the malondialdehyde and reactive oxygen species levels in cortex of depressed mice.SFSP played a role in preventing depression by increasing the absorption of aromatic amino acids,increasing monoamine neurotransmitters concentrations in hippocampus,and preventing oxidative stress in cortex.
