青海云杉无性系的遗传多样性

摘要 【目的】通过对青海云杉无性系遗传多样性和亲缘关系的研究,以期为青海云杉高世代种子园的建立提供可靠的建园材料.【方法】以109个青海云杉种子园内的无性系作为研究对象,采用正交试验设计和单因素分析方法,建立并优化了青海云杉的SSR-PCR反应体系,利用筛选出的10对有效引物,分析青海云杉无性系遗传多样性.【结果】基于SSR分子标记对109个青海云杉无性系遗传多样性分析,共扩增出230条多态性条带,多态百分率(PPL)为100.00%,平均有效等位基因数(n e)为14.76,平均观测杂合度(H o)为0.06,平均期望杂合度(H e)为0.92,平均Shannon′s多态性信息指数(I)为2.77.【结论】采用UPGMA聚类分析方法,将109个无性系分为5组,差异明显,此结果与种子园建园材料的地理来源广泛和个体间基因交流广泛吻合,揭示了青海云杉无性系间的遗传多样性较为丰富. 【Objective】Through the study of genetic diversity and kinship of Picea crassifolia clone,we hope to provide reliable construction for the establishment of the seed orchard of Picea crassifolia in higher generation garden materials.【Method】This dissertation conducts research on clones from 109 seed gardens of Picea crassifolia,using orthogonal experiment design and single factor analysis method,to establish and optimize the Picea crassifolia SSR-PCR reaction system.By screening 10 pairs of effective primers,the genetic diversity and genetic relationship of Qinghai spruce clones are analyzed.【Result】The result shows that 230 polymorphic bands are amplified based on SSR molecular markers for the genetic diversity of 109 clones of Picea crassifolia.The percentage of polymorphism(PPL)is 100.00%,and the average number of effective alleles(ne)is 14.76,the average observed heterozygosity(Ho)is 0.06,the average expected heterozygosity(He)is 0.92,and the average Shannon′s polymorphism information index(I)is 2.77.【Conclusion】Using UPGMA cluster analysis method,109 clones are divided into 5 groups,which are obviously differentiated,and the results are consistent with the extensive geographical sources of seed orchard materials and the extensive gene exchange between individuals,revealing that the genetic diversity among spruce clones in Qinghai is relatively rich.
