摘要
以超声辅助法提取藜麦糠黄酮粗提物为原料,采用大孔树脂进行吸附与解吸附。考察了7种树脂对藜麦糠黄酮的吸附与解吸附性能,借助静态吸附动力学实验筛选最优纯化藜麦糠黄酮的树脂。结合静态与动态吸附解吸实验得出最优纯化条件:样液浓度0.5mg/mL,上样量35mL,上样液pH 6.0,乙醇浓度75%,乙醇溶液体积180mL。在此条件下分离纯化后,藜麦糠中黄酮纯度从13.0%增加到68.0%,回收率为77.4%。表明用HPD950大孔树脂对藜麦糠黄酮分离纯化有良好的效果。对纯化后的藜麦糠黄酮样品进行高效液相色谱分析,样品中槲皮素、槲皮苷、芦丁和异槲皮素4种成分得到确认。
The flavonoids crude extracts of quinoa chaff extracted by ultrasonic-assisted method are as the raw material,and the macroporous resin is used for adsorption and desorption.The adsorption and desorption properties of seven kinds of resins on quinoa chaff flavonoids are investigated.The optimum resin for purification of quinoa chaff flavonoids is screened by static adsorption kinetics experiment.The optimal purification conditions are obtained by combining static and dynamic adsorption-desorption experiments:sample concentration is 0.5 mg/mL,sample loading quantity is 35 mL,sample solution pH is 6.0,ethanol concentration is 75%,ethanol solution volume is 180 mL.After separation and purification under these conditions,the purity of flavonoids in quinoa chaff changes from 13.0%to 68.0%,and the recovery rate is 77.4%.The results show that HPD950 macroporous resin has a good effect on the separation and purification of quinoa chaff flavonoids.Four components of quercetin,quercetin,rutin and isoquercetin in the samples of quinoa chaff flavonoids are identified by high performance liquid chromatography.
