壮药拔毒生肌膏对糖尿病大鼠创面愈合及表皮生长因子、血管内皮生长因子、溶菌酶表达水平的影响 被引量:4

摘要 目的研究壮药拔毒生肌膏对糖尿病大鼠创面愈合的促进作用及其可能机制。方法将96只大鼠随机分为假手术组、模型组、贝复济组、拔毒生肌膏组各24只。除假手术组外,其他组均通过腹腔注射链脲佐菌素建立糖尿病大鼠模型。模型制备成功1周后,对各组大鼠建立创面模型;24 h后模型组与假手术组均外敷生理盐水纱条,贝复济组、拔毒生肌膏组分别外敷贝复济纱条及拔毒生肌膏纱条。分别在干预后3 d、6 d、12 d,计算各组大鼠创面愈合率,测定血清溶菌酶水平、血清和组织表皮生长因子(EGF)、血管内皮生长因子(VEGF)水平,并于光镜下观察大鼠创面形态变化。结果 (1)假手术组创面愈合快速,而模型组干预后3 d及12 d的创面愈合率均低于假手术组(均P<0.05)。拔毒生肌膏组、贝复济组创面组织水肿和出血较早消失,且肉芽组织较快生长;与模型组比较,贝复济组干预后3 d及6 d的创面愈合率均升高,拔毒生肌膏组干预后6 d及12 d的创面愈合率均升高(均P<0.05)。拔毒生肌膏组各时间点的创面愈合率与假手术组比较差异均无统计学意义(均P>0.05),而贝复济组干预后12 d的创面愈合率低于假手术组和拔毒生肌膏组(均P<0.05)。(2)贝复济组、拔毒生肌膏组干预后6 d、12 d的血清及组织EGF、VEGF水平,以及各个时间点的血清溶菌酶水平均高于模型组(均P<0.05)。干预后6 d拔毒生肌膏组的血清EGF水平低于贝复济组(P<0.05),两组各时间点血清VEGF及组织EGF和VEGF、血清溶菌酶水平比较差异均无统计学意义(均P>0.05)。结论拔毒生肌膏可能通过调控血清和组织中的EGF、VEGF及血清溶菌酶表达,改善创面组织水肿,促进肉芽组织生长,促进糖尿病大鼠创面愈合。 Objective To study the facilitation of Zhuang medicine Badu Shengji Ointment on wound healing in diabetic rats and its possible mechanism. Methods Ninety-six rats were randomized to sham operation group, model group, Beifuji group and Badu Shengji Ointment group, with 24 rats in each group. Except the sham operation group, the other groups established a rat model of diabetes mellitus by intraperitoneal injection of streptozotocin. One week after successful modeling, wound model was established in all groups;twenty-four hours later, gauze soaked with normal saline was applied externally to the model group and the sham operation group, while gauze soaked with Beifiji and Badu Shengji Ointment was applied externally to the Beifuji group and the Badu Shengji Ointment group, respectively. Three, 6 and 12 days after intervention, the wound healing rate of each group was calculated, serum lysozyme level, and the levels of epidermal growth factor(EGF) and vascular endothelial growth factor(VEGF) in serum and wound tissue were determined, and the morphological changes of rat wound tissue were observed under light microscope. Results(1) Rapid wound healing was observed in the sham operation group, whereas the model group exhibited lower wound healing rates three and 12 days after intervention as compared with the sham operation group(all P<0.05). In the Badu Shengji Ointment group and the Beifuji group, wound tissue edema and hemorrhage disappeared in early stage, and the growth of granular tissue was fast;compared with the model group, the Beifuji group yielded higher wound healing rates three and six days after intervention, while the Badu Shengji Ointment group yielded higher wound healing rates six and 12 days after intervention(all P<0.05). There was no statistically significant difference in wound healing rate at each time point between the Badu Shengji Ointment group and the sham operation group(all P>0.05), but the wound healing rate 12 days after intervention in the Beifuji group was lower than the rate in the sha
