摘要
目的:探讨温阳化瘀方对5/6肾切除大鼠Sirt/NF-κB炎症通路的调节作用。方法:SD大鼠32只,随机分为假手术组、模型组、温阳化瘀方组、西药组,每组8只。温阳化瘀方组予以温阳化瘀方水煎液15.1 g/(kg·d)灌胃,西药组予以洛汀新1mg/(kg·d)灌胃,连续灌胃4周;全自动生化仪检测各组大鼠血SCr、尿NAG、24 h尿蛋白水平,ELISA检测血清核因子κB(NF-κB)和人单核细胞趋化蛋白-1(MCP-1)水平,qRT-PCR检测大鼠肾组织沉默信息调节因子2相关酶1(SIRT1)mRNA表达。结果:与模型组比较,温阳化瘀方组、西药组大鼠血SCr、尿NAG水平均显著降低(P<0.01),NF-κB和MCP-1水平均显著降低(P<0.01);温阳化瘀方组SIRT1mRNA表达上升(P<0.05),西药组SIRT1mRNA表达差异无统计学意义(P>0.05)。结论:温阳化瘀方能上调SIRT水平,抑制NF-κB和MCP-1表达,改善肾功能。
Objective:To investigate the regulatory effect of Wenyang Huayu Recipe on Sirt/NF-kappa B inflammatory pathway in 5/6 nephrectomized rats.Methods:32 SD rats were randomly divided into four groups:sham operation group,model group,Wenyang Huayu Recipe group,Western medicine group with 8 rats in each group.The Wenyang Huayu Recipe group was given water decoction of Wenyang Huayu Recipe 15.1 g/(kg·d),the Western medicine group was given Lotinxin 1 mg/(kg·d),each group was intervened for 4 weeks.The levels of SCr,NAG,24-hour urinary protein were determined by automatic biochemical analyzer,serum NF-kappa B and MCP-1 were tested by ELISA,the expressions of SIRT1 mRNA in kidney were detected by qRT-PCR.Results:The levels of SCr and NAG in Wenyang Huayu Recipe group,Western medicine group were significantly lower than those in the model group(P<0.01).The levels of serum NF-kappa B and MCP-1 in the Wenyang Huayu Recipe group,the Western medicine group were significantly lower than those in the model group(P<0.01).The expression of SIRT1 mRNA in Wenyang Huayu Recipe group was increased significantly(P<0.01),compared with the model group,the difference was significant(P<0.05).Conclusion:Wenyang Huayu Recipe can up-regulate SIRT expression,inhibit the expression of NF-kappa B and MCP-1,improve renal function.
