Study on the kidney-tonifying effect and mechanism of Shenbao Tablets

摘要 目的研究肾宝片的补肾作用,并探讨其作用机制。方法采用腺嘌呤致Wistar大鼠肾阳虚模型,按体重随机分为正常对照组,模型对照组,肾宝片1.14、0.57、0.29 g/kg剂量组,枸橼酸西地那非片组,每组13只。除正常对照组外,其余大鼠灌胃腺嘌呤500 mg/kg体重,连续10 d。造模第二天开始各组分别灌胃给药,正常对照组和模型对照组灌胃相同容积的蒸馏水,连续给药24 d。比较各组大鼠体重、自主活动次数、低温游泳存活时间、血清睾酮(T)、血尿素氮(BUN)、肌酐(Cre)、超氧化物歧化酶(SOD)、丙二醛(MDA)、环磷酸鸟苷(cGMP)及总一氧化氮合酶(NOS)水平。采用去势大鼠模型,按体重随机选取12只为正常对照组,其余大鼠进行去势手术,将去势大鼠按体重随机分为模型对照组、肾宝片2.28、1.14、0.57 g/kg剂量组及枸橼酸西地那非片组,每组12只。各组分别灌胃给药,1次/d,连续给药24 d,正常对照组和模型对照组灌胃相同容积的蒸馏水。比较各组勃起潜伏期、阴茎海绵体内压/血压(ICP/BP)、cGMP和总NOS及血清T水平。结果肾阳虚模型对照组体重、自主活动次数、低温游泳存活时间、血清T含量、阴茎海绵体内cGMP水平低于正常对照组,血清BUN、Cre、MDA含量高于正常对照组(P <0.05)。肾宝片1.14、0.57、0.29 g/kg剂量组低温游泳存活时间长于模型对照组、血清BUN含量低于模型对照组,肾宝片1.14、0.57 g/kg剂量组自主活动次数、阴茎海绵体内cGMP水平高于模型对照组,肾宝片0.57 g/kg剂量组血清T含量、SOD活性高于模型对照组、Cre含量低于模型对照组(均P <0.05);肾宝片1.14、0.57、0.29 g/kg剂量组体重、血清MDA含量、阴茎海绵体内总NOS水平与模型对照组比较,差异无统计学意义(P> 0.05)。去势大鼠模型对照组勃起潜伏期高于正常对照组,血清T含量、阴茎海绵体ICP值、ICP/BP比值、阴茎海绵体内cGMP水平低于正常对照组(P <0.01) Objective To study the kidney-tonifying effect of Shenbao Tablets and explore its mechanism.Methods The model of kidney-yang deficiency of Wistar rats induced by adenine was adopted.They were randomly divided into normal control group,model control group,Shenbao Tablets 1.14,0.57 and 0.29 g/kg dose groups,Sildenafil Citrate Tablets group,with 13 rats in each group.Except the normal control group,the other rats were gavaged with adenosine 500 mg/kg for 10 d.On the second day of modeling,each group was administrated by gavage for 24 d.The body weight,the number of independent activities,the survival time of swimming at low temperature,the serum testosterone(T),blood urea nitrogen(BUN),creatinine(Cre),serum superoxide dismutase(SOD),malondialdehyde(MDA),cyclic guanosine phosphate(cGMP)and total nitric oxide synthase(NOS)were compared in each group.The castrated rat model was used,and 12 rats were randomly selected according to the body weight as the normal control group.The remaining rats were castrated.The castrated rats were randomly divided into model control group,Shenbao Tablets 2.28,1.14 and 0.57 g/kg dose groups,and Sildenafil Citrate Tablets group,with 12 rats in each group.Each group was administrated by gavage once a day for 24 d,normal control group and model control group were gavaged the same volume of distilled water.The erectile latency,intracavernous pressure/blood pressure(ICP/BP),cGMP and total NOS levels and serum T levels were compared in each group.Results The body weight,the number of independent activities,the survival time of low temperature swimming,serum T levels and cGMP in penile sponge in model control group of kidney-yang deficiency were lower than those in normal control group,and serum BUN,Cre and MDA levels were higher than those in normal control group(P<0.05).The survival time of low temperature swimming of Shenbao Tablets 1.14,0.57 and 0.29 g/kg dose groups were longer than those of model control group,the BUN content was lower than that of model control group,the number of indepe
