摘要
为监测猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)基因变异情况,对广东地区某猪场患有猪繁殖与呼吸综合征(PRRS)仔猪的肺脏样品进行检测及病毒分离,在猪肺泡巨噬细胞(PAM)上成功分离1株病毒并命名为GDjm株,然而分离的毒株不能感染Marc-145细胞。全基因组进化分析、同源性比对及重组分析结果显示,GDjm属于重组毒株,与GDZS2016的同源性为96.2%,与中国高致病性毒株HUN4同源性为96.0%,与美洲型经典毒株VR-2332的同源性为87.4%,与NADC30同源性为82%,与欧洲型毒株Lelystad-virus同源性为57.9%。NSP2序列比对和同源性分析结果显示,GDjm与高致病性毒株HUN4、JXA1、JXwn06相似,在511、534~562位存在氨基酸缺失。GP5序列比对以及同源性分析结果显示,GDjm毒株在GP5抗原表位存在氨基酸突变。经重组分析得出GDjm株为高致病性毒株HUN4与华南地区流行毒株GDZS2016的重组毒株,重组区域为6723~9521 bp、11998~15019 bp。
To understand the evolution of the epidemic strains of the porcine reproductive and respiratory syndrome virus(PRRSV),the lung samples were collected of pigs suspected of having porcine reproductive and respiratory syndrome in a pig farm in Jiangmen City,Guangdong Province.The samples were tested and proved to be positive.The samples were subjected to virus isolation on porcine alveolar macrophages,and the successfully obtained strain was designated as the GDjm strain.However,the isolated strain was unable to infect Marc-145 cells.Genome-wide evolution analysis,homology alignment and recombinant analysis showed that GDjm was a recombinant strain,in 96.2%homology with GDZS2016 and 96.0%homology with the highly pathogenic Chinese strain HuN4,in 87.4%homology with the American classic strain VR-2332,82%homology with NADC30,and in a low 57.9%homology with the typical European strain the Lelystad-virus.Sequence alignment and homology analysis of NSP2 showed that GDjm was similar to the highly pathogenic strains HUN4,JXA1,and JXwn06,with amino acids deleted at positions 511,534 to 562.GP5 sequence alignment and homology analysis showed that GDjm presented amino acid mutation at GP5 epitope.The recombination analysis showed that the GDjm strain was a highly pathogenic strain HUN4 and a variant strain GDZS2016 in south China.The recombinant regions were 6723-9521 bp and 11998-15019 bp.
