共聚焦超分辨率成像与SIM超分辨率成像在蒿草生物组织样本荧光成像中的使用价值分析

摘要 目的研究分析共聚焦超分辨率成像与SIM超分辨率成像在蒿草生物组织样本荧光成像中的使用价值。方法使用激光扫描共聚焦显微镜对蒿草生物组织样本进行荧光成像和使用结构光照明显微镜对蒿草生物组织样本进行SIM超分辨率荧光成像。实验分为三组。(1)使用激光扫描共聚焦显微镜对蒿草组织样本共聚焦普通成像;(2)使用激光扫描共聚焦显微镜的超分辨率成像模块对蒿草组织样本超分辨率成像;(3)使用结构光照明显微镜对蒿草组织样本SIM超分辨率成像。结果激光扫描共聚焦显微镜不仅可获取蒿草组织样本的不同倍数共聚焦普通图像,而且可获取蒿草组织样本的共聚焦超分辨率图像;结构光照明显微镜可获取蒿草组织样本的SIM超分辨率荧光图像。结论激光扫描共聚焦显微镜和结构光照明显微镜均可对生物组织样本进行超分辨率成像。共聚焦超分辨率成像图像清晰度较高,色彩对比度明显,而SIM超分辨率成像具有显示组织样本局部的超微结构细节更加突出的特点。 Objective To study the value of confocal super-resolution imaging and SIM super-resolution imaging in fluorescence imaging of wormwood biological tissue samples.Methods The laser scanning confocal microscope was used for fluorescence imaging of wormwood biological tissue samples,and the structure-light illumination microscope was used for SIM super-resolution fluorescence imaging of wormwood biological tissue samples.This study was divided into three groups.(1)Confocal common imaging of wormwood tissue samples was performed using laser scanning confocal microscope;(2)Super-resolution imaging of wormwood tissue samples was performed using super-resolution imaging module of laser scanning confocal microscope;(3)Wormwood tissue sample SIM superresolution imaging was performed using structured light illumination microscope.Results The laser scanning confocal microscope could not only obtain common images of different multiples of wormwood tissue samples,but also confocal super-resolution images of wormwood tissue samples;structured light illumination microscope could obtain SIM super-resolution fluorescence image of wormwood tissue samples.Conclusion Both laser scanning confocal microscope and structured light illumination microscope can perform super-resolution imaging of biological tissue samples.Confocal super-resolution imaging has higher resolution and obvious color contrast,while SIM super-resolution imaging has the characteristics of showing the local ultra-structural details of tissue samples more prominently.
