芍药种胚启动培养及丛生芽诱导研究 被引量:4

摘要 为探究芍药种胚启动培养、丛生芽诱导及增殖过程中的主要影响因素,以芍药种胚为外植体,研究不同发育阶段的种胚(授粉后65,75,85,95d)、暗培养时间(0,4,8d)以及3种激素(1.0mg·L^-16-BA、0.5mg·L^-1GA3、0.2mg·L^-1NAA)对芍药种胚启动培养的影响;同时研究6-BA(0.5mg·L^-1和1.0mg·L^-1)与GA3(0.5mg·L^-1和1.0mg·L^-1)的激素组合对种胚丛生芽初步诱导的影响;也探究不同浓度的6-BA(1.0,2.0,3.0,4.0mg·L^-1)与1.0mg·L^-1GA3组合对种胚丛生芽二次诱导以及不同浓度的CH(0,0.3,0.5g·L^-1)对种胚丛生芽增殖成苗的影响;本试验还以子叶节为外植体,接种在种胚丛生芽最适的诱导及增殖的培养基中,对种胚丛生芽及子叶节丛生芽形态特征的差异进行初步探究。结果表明:芍药种胚启动培养的最佳胚龄为授粉后的75d;胚苗最佳的培养方式为暗培养4d后置于正常光照条件下培养;启动培养最适激素组合为0.5mg·L^-1GA3+1.0mg·L^-16-BA;丛生芽初步诱导及二次诱导最适培养基分别为MS+1.0mg·L^-1GA3+1.0mg·L^-16-BA及MS+1.0mg·L^-1GA3+2.0mg·L^-16-BA;丛生芽增殖的最适培养基为1.0mg·L^-1MS+GA3+3.0mg·L^-16-BA+0.3g·L^-1CH+0.1mg·L^-1NAA+1.0g·L^-1PVP,适宜浓度的CH可显著提高丛生芽的增殖系数,也可以使丛生芽苗更健壮。丛生芽初步诱导过程中,子叶节的诱导效果及生长状态优于种胚直接诱导,但种胚具有更高的萌发潜质。种胚丛生芽的增殖效果优于子叶节丛生芽。 The test is intended to explore the Paeonia lactiflora embryo start training and main influence factors in the process of multiple shoots induction and proliferation,among them with peony embryo as explant,studied the different development stage of embryo(65,75,85,95d after pollination),dark incubation time(0,4,8d)and three kinds of hormone(1.0mg·L^-16-BA,0.5mg·L^-1GA3,0.2mg·L^-1NAA)of Herbaceous peony embryo start training;The effects of hormone combinations of 6-BA(0.5mg·L^-1 and 1.0mg·L^-1)and GA3(0.5 mg·L^-1 and 1.0mg·L^-1)on embryo multiple shoots induction were studied.The effects of 6-BA(1.0,2.0,3.0,4.0mg·L^-1)and 1.0mg·L^-1GA3 at different concentrations on secondary induction of embryo multiple shoots and CH(0,0.3,0.5mg·L^-1)at different concentrations on embryo multiple shoots proliferation into seedlings were also investigated.In this study,cotyledon nodes were used as explants and inoculated in the optimal medium for induction and proliferation of embryo multiple shoots,and the morphological characteristics of embryo multiple shoots and cotyledon node multiple shoots were preliminarily investigated,explore effects of herbaceous peony embryo start training and the main factors of multiple shoots of inducing proliferation.The optimal embryo age of the starting culture of the peony embryos was 75 days after pollination.The best embryo culture method was dark culture for 4 days and then placed under normal light conditions;the most suitable hormone combination was 0.5mg·L^-1GA3+1.0mg·L^-16-BA;the most suitable medium for initial induction was MS+1.0mg·L^-1GA3+1.0mg·L^-16-BA;the most suitable medium for secondary induction was MS+1.0mg·L^-1GA3+2.0mg·L^-16-BA;the most suitable medium for multiple shoots proliferation was MS+1.0mg·L^-1GA3+3.0mg·L^-16-BA+0.3g·L^-1CH+1.0g·L^-1PVP.The appropriate concentration of CH could significantly improve the proliferation coefficient of clusters and also make multiple shoots more robust.During the initial induction of multiple shoots,the cotyledon node
