摘要
目的研究不同剂量枸杞多糖(LBP)对高糖诱导的RSC96雪旺细胞增殖的影响。方法分别采用不同葡萄糖浓度(12.5、25、50、100 mmol/L)的杜氏改良Eagle培养基(DMEM)培养大鼠RSC9648h,用四唑盐比色法(MTT)检测不同葡萄糖浓度对RSC96细胞活力的影响。确定下一步实验的正常对照组及高糖组的葡萄糖浓度,在此基础上给予不同剂量(100、200、400、800μg/mL)LBP培养48 h,用MTT法检测不同剂量LBP对RSC96细胞增殖的影响。结果在不同浓度的葡萄糖中,25 mmol/L葡萄糖浓度组RSC96细胞活力最高,100 mmol/L组RSC96细胞活力下降最为显著;与高糖组比较,200、400、800μg/mL的LBP均有促进RSC96细胞增殖的作用,LBP为400μg/mL作用最明显(P<0.05)。结论LBP为400μg/mL对高糖诱导的RSC96细胞增殖作用最佳。
Objective To study the effects of different doses of Lycium barbarum polysaccharides(LBP) on the proliferation of RSC96 Schwann cells induced by high glucose.Methods RSC96 cells were cultured respectively in Dulbecco’s modified eagle medium(DMEM) with different concentration of glucose(12.5,25,50,100 mmol/L) for 48 h. The activity of cells were detected by MTT. On this basis,the different doses of LBP(100,200,400,800 μg/mL) were used to culture RSC96 cells for 48 hours,and the proliferation of RSC96 cell was detected by MTT method. Results In different concentrations of glucose,the activity of RSC96 Schwann cells in 25 mmol/L glucose group was the highest,and the greatest reduction of activity of RSC96 Schwann cells was in the 100 mmol/L glucose.Compared with the high glucose group(100 mmol/L),the concentrations of LBP(200,400,800 μg/mL) promoted the proliferation of RSC96 Schwann cells induced by high glucose,the proliferation activity of RSC96 Schwann cells in LBP(400 μg/mL)increased significantly(P<0.05).Conclusion LBP(400 μg/mL) has the best effect on the proliferation of RSC96 cells induced by high glucose.
