益气养阴活血方对2型糖尿病大鼠PI3K/Akt信号通路相关蛋白表达的影响 被引量:7

Effect of Yiqi Yangyin Huoxue Prescription on Expression of PI3K/Akt Signaling Pathway Related Protein in Type 2 Diabetic Rats

摘要 目的:探讨益气养阴活血方对2型糖尿病(type 2 diabetes mellitus,T2DM)大鼠胰岛β细胞功能及胰岛细胞中磷酸化磷脂酰肌醇3激酶(phosphatidylinositol-3-kinasem,PI3K)、磷酸肌醇依赖性蛋白激酶1(phosphoinositide-dependent protein kinase 1,PDK1)蛋白表达的影响。方法:随机选取10只雄性SD大鼠作为正常对照组,其余50只为造模组。正常对照组给予普通饲料加生理盐水灌胃,通过高脂饮食加小剂量腹腔注射链脲佐菌素建立T2DM大鼠模型并随机分为模型组、津力达组、益气养阴活血方高、中、低剂量组,分别给予生理盐水、津力达颗粒(1.5g/kg)、益气养阴活血方高、中、低剂量(55.00 g/kg,27.50g/kg、13.75g/kg)灌胃,每日1次,在第28天测定空腹血糖和空腹胰岛素水平并计算胰岛素抵抗指数和胰岛素分泌指数,分别采用免疫组化法和免疫蛋白印迹法检测胰岛PI3K、PDK1的蛋白表达。结果:与模型组比较,益气养阴活血方各剂量组均能明显降低糖尿病大鼠空腹血糖、胰岛素抵抗指数水平,升高空腹胰岛素、胰岛素分泌指数、PI3K、PDK1水平。结论:益气养阴活血方能够显著改善T2DM大鼠胰岛β细胞功能,其改善作用可能是通过上调PI3K/Akt信号通路PI3K和PDK1蛋白表达来实现的。 Objective:To investigate the effects of Yiqi Yangyin Huoxue Prescription(YYHP)on the function of islet beta cells and expression of phosphatidylinositol-3-kinasem(PI3K)and phosphoinositide-dependent protein kinasem 1(PDK1)proteins in rats with type 2 diabetes mellitus(T2DM).Methods:10 male Sprague Dawley rats were randomly selected as normal control group,and other 50 rats were prepared for models.Rats in the normal control group were fed with ordinary diet.In addition,The T2DM rat models were established by high-fat diet and low-dose intraperitoneal injection of streptozotocin.Then the 50 T2DM rat models were randomly divided into model group,Jinlida group,and YYHP with high-does,mid-does,low-does group.The rat models in corresponding groups were respectively given saline,Jinlida granules(1.5g/kg),high-does(55.00 g/kg),mid-does(27.50g/kg),low-does(13.75g/kg)of YYHF once a day for 28 days.After the experiment,the level of fasting blood glucose(FBG)and fasting insulin(FINS)were tested and the homeostasis model assessment-insulin resistance(HOMA-IR)and indexes of insulin secretion(HOMA-β)were calculated.Moreover,The protein expression levels of PI3K and PDK1 in pancreatic tissues were detected by immunohistochemistry and western blot.Results:Compared with the model group,the levels of FBG and HOMA-IR in each does of YYHF group were decreased obviously,and FINS,HOMA-β,PI3K and PDK1 were increased obviously.Conclusion:Yiqi Yangyin Huoxue Prescription can significantly improve the function of β cells in T2DM rats,and its improvement may be achieved by up-regulating the protein expression of PI3K and PDK1 on PI3K/Akt signaling pathway.
