摘要
目的研究生姜压榨汁对脑缺血再灌注大鼠脑组织缺氧诱导因子1α(hypoxia-inducible factor 1α,HIF1α)蛋白和Na^+/H^+交换蛋白1型(Na^+/H^+exchanger protein 1,NHE1)表达水平的影响。方法通过线栓法构建大鼠大脑中动脉缺血再灌注模型,造模成功的大鼠随机分为模型组(MCAO)、生姜压榨汁高(H)、中(M)和低(L)剂量组,另设假手术组(CLT)为对照,各组大鼠经灌胃给药5 d,神经功能评分后,病理组织检查大脑病变情况,干湿重法测大脑含水量,免疫荧光法检测NHE1蛋白在脑组织中的表达,Western Blot法研究HIF-1α蛋白和NHE1蛋白水平。结果与CTL组比较,MCAO组大鼠出现明显的神经功能障碍,病理检测结果显示脑组织出现明显的梗死灶,脑组织含水量明显升高,NHE1蛋白和HIF-1α蛋白表达水平明显升高(P<0.05)。与MCAO组相比,生姜H、M剂量组能明显改善MCAO大鼠神经病学症状,显著缩小脑梗死面积,降低脑组织含水量,并降低NHE1蛋白和HIF-1α蛋白表达水平(P<0.05)。结论生姜对脑缺血再灌注大鼠脑组织可能发挥保护作用。
Objective To study the effects of Shengjiang(Zingiber officinale)on levels of hypoxia-inducible factor 1α(HIF1α)protein and Na^+/H^+exchanger protein 1(NHE1)in brain tissues of cerebral ischemia-reperfusion rats.Methods The model of focal cerebral ischemia in rats was established by the right middle cerebral artery occlusion(MCAO)and reperfusion.Male SD rats were randomly divided into MCAO group,high(H),middle(M),low(L)dosage groups of Shengjiang(Zingiber officinale),and sham-operation group was constructed as control(CTL).After 5 days,the neurological outcomes were scored.Rats in each group were respectively killed to obtain their brain tissue for HE staining.The changes of brain water content were measured by the wet and dry weight methods.The expression of NHE1 protein in brain tissue was detected by immunofluorescence.The levels of HIF-1αprotein and NHE1 protein were studied by Western Blot method.Results Compared with CTL group,the MCAO group showed obvious neurological dysfunction.Pathological examination results showed that there was obvious infarction in brain tissue,water content in brain tissue increased significantly,and the levels of NHE1 protein and HIF-1αprotein increased significantly(P<0.05).High and middle dosage of Shengjiang(Zingiber officinale)could obviously lessen the pathological changes,neurological function scores,and decrease brain water content and the expression levels of NHE1 protein and HIF-1αprotein compared with the MCAO group.Conclusion Shengjiang(Zingiber officinale)may play a protective role in brain tissues of cerebral ischemia-reperfusion rats.
