摘要
3-羟基-3-甲基戊二酰辅酶A还原酶(HMGR)是植物萜类化合物生物合成的关键酶,在茅苍术挥发油生物合成过程中起重要作用,但目前茅苍术AlHMGR全长序列尚不清楚。本研究在前期茅苍术转录组学研究的基础上,筛选AlHMGR序列,并进行其c DNA序列的全长克隆,然后分析AlHMGR的序列特征及其编码蛋白的结构特征。结果显示,所得两条AlHMGR全长cDNA序列分别为1749 bp和1770 bp,分别编码582个和589个氨基酸;系统进化分析表明与菊科植物朝鲜蓟亲缘关系最近。本研究还探讨了AlHMGR编码蛋白的氨基酸序列特征、蛋白结构特征及亚细胞定位。本研究结果为进一步阐明茅苍术萜类化合物生物合成和调控的分子机制提供了理论依据。
3-hydroxy-3-methyl glutaryl coenzyme A reductase(HMGR),a key enzyme of terpene biosynthesis,plays an import role in essential oil biosynthesis in Atractylodes lancea.However,the full sequence of AlHMGR has not been achieved till now.In this study we screened and cloned two AlHMGR genes based on the previous results of A.lancea transcriptome data,and analyzed the characteristic of their gene sequences and encoding protein sequences.The results showed that the full length of two AlHMGR genes was 1749 bp and 1770 bp encoding 582 and 589 amino acids,respectively.Phylogenetic analysis suggested the highest homology with Cynara cardunculus var.scolymus.The characteristic of AlHMGRs encoding protein sequences,structural feature,and subcellular localization were also predicted.The results will facilitate the elucidation and regulation of terpene biosynthesis in A.lancea.
