NLRP3炎性小体基因多态性与慢性牙周炎易感性的相关性研究 被引量:7

摘要 目的评估NLRP3(rs4612666)单核苷酸多态性(SNP)是否与中国人群慢性牙周炎(CP)的发病有关。方法对236例中重度非吸烟CP患者和191例牙周健康对照者(HC)采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)法用于鉴定NLRP3(rs4612666)基因型。结果与对照组比较,CP患者中观察到NLRP3(rs4612666)SNP的CT、TT基因型及T等位基因与增高的CP发病风险相关(P均<0.01)。经过按性别和CP严重程度分层分析,男性人群携带NLRP3(rs4612666)CT、TT基因型和T等位基因个体CP的发病风险增高2~3倍。而在女性组及中度和重度CP分组中均未观察到此相关性。结论NLRP3(rs4612666)SNP的CT和TT基因型可能在男性CP的致病途径中发挥作用。未来有必要在独立的大样本人群中对结果进行重复研究以验证rs4612666 SNP和CP的相关性。 Objective To evaluate whether NLRP3(rs4612666)single nucleotide polymorphism(SNP)is related to the susceptibility of chronic periodontitis(CP)in Chinese han population.Methods A total of 236 moderate to severe nonsmoking CP patients and 191 periodontal health controls were included in the study.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method was used to identify the NLRP3(rs4612666)genotype.Results Compared with the control group,CT,TT genotype and T allele of NLRP3(rs4612666)were observed to be associated with increased CP risk in CP patients(all P<0.01).After stratified analysis by sex and CP severity,the risk of CP in individuals with NLRP3(rs4612666)CT,TT genotype and T allele was 2 to 3 times higher in the male population.This association was not observed in the female group or in the moderate and severe CP groups.Conclusion Both CT and TT genotypes of NLRP3(rs4612666)SNP may play a role in the pathogenesis of CP in males.Replication studies of our results in independent large populations are necessary to validate these associations in diverse populations.
