摘要
目的:建立测定利咽饮中绿原酸含量的HPLC方法。方法:选用Diamonsil C18(250mm×4.6mm,5μm)色谱柱测定绿原酸含量,流动相为乙腈-0.4%磷酸水溶液(9∶91),检测波长为327nm,柱温为30℃,流速为1mL/min。结果:在该色谱条件下,样品中的绿原酸分离效果理想,绿原酸在9.28~55.68μg/mL浓度范围内与峰面积呈良好的线性关系。回归方程为A=19.27C+34.52(R2=0.999 0),加样平均回收率为99.06%,RSD=0.51%(n=6)。结论:建立的方法快速准确,简便灵敏,重复性好,结果可靠,可用于利咽饮中绿原酸的含量测定,为控制利咽饮的质量提供参考。
Objective:To establish an HPLC method to detemine the content of chlorogenic acid in Liyanyin.Methods:The separation was performed on the Diamonsil C18(250 mm×4.6 mm,5μm)column with acetonitrile-0.4%phosphate 9∶91 as the mobile phase,the wavelength of UV detector was 327 nm and the column temperature was 30℃.The flow rate was 1 mL·min-1.Results:Under this chromatographic condition,the separation effect of chlorogenic acid in the sample is ideal.The amount of chlorogenic acid injected into the sample ranges from 9.28 to 55.68 ug/mL and the peak area shows a good linear relationship.The regression equation was A=19.27 C+34.52(R=0.9995)and the average recovery rate was 99.06%with RSD of 0.51%(n=6).Conclusion:The method is rapid,accurate,simple,sensitive,reproducible and reliable.It can be used to determine the content of chlorogenic acid in Liyan drink and provide a reliable method for controlling the quality of Liyan drink.
