电针对糖尿病肌萎缩大鼠骨骼肌及血糖的影响 被引量:10

Effects of electroacupuncture on skeletal muscle and blood glucose in rats with diabetic amyotrophy

摘要 目的:探究电针对糖尿病肌萎缩大鼠骨骼肌及血糖的作用。方法:从40只SD大鼠中随机选取10只作为对照组,不进行任何干预;其余30只采用腹腔注射链脲佐菌素(STZ)60 mg/kg制备糖尿病(DM)模型,再运用右后肢血管结扎术制备肌萎缩模型,将造模成功大鼠随机分为模型组和电针组,各10只(排除造模不成功及死亡大鼠10只)。选取右侧"胰俞""肾俞""足三里"及"三阴交"对电针组大鼠进行电针(疏密波,2 Hz/15 Hz)干预,每次20 min,每日1次,持续3周。电针干预前后,目内眦取血并采用"葡萄糖氧化酶-过氧化物酶"法检测各组大鼠空腹血糖水平,ELISA法检测血清胰岛素含量;干预结束后,HE染色法观察各组大鼠右后肢缺血骨骼肌肌肉形态,实时定量PCR(real-time PCR)法检测右后肢缺血骨骼肌肌肉组织中骨骼肌特异性泛素连接酶E3--肌肉萎缩盒F(MAFbx)及肌肉环状指-1(Mu RF1),叉头框转录因子O3a(FOXO3a)m RNA的表达。结果:干预前,模型组与电针组大鼠体质量均低于对照组(P<0.01);干预后,对照组大鼠体质量升高,模型组和电针组大鼠体质量均降低(P<0.01)。与对照组比较,模型组大鼠空腹血糖显著升高,胰岛素含量明显减少(P<0.01);与模型组比较,电针组大鼠干预后空腹血糖明显降低,胰岛素含量显著增大(P<0.05)。模型组大鼠肌纤维明显断裂,细胞核减少,出现核固缩甚至溶解;电针组大鼠干预后肌肉组织形态较模型组有所改善。与对照组比较,模型组大鼠右后肢缺血骨骼肌肌细胞横截面积减小(P<0.01);与模型组比较,电针组大鼠干预后右后肢缺血骨骼肌肌细胞横截面积增大(P<0.05)。与对照组比较,模型组大鼠右后肢缺血骨骼肌肌肉组织中泛素连接酶E3相关基因MAFbx和Mu RF1,FOXO3am RNA的水平均显著增加(P<0.01,P<0.05);与模型组比较,电针组大鼠干预后MAFbx和Mu RF1,FOXO3a m RNA的水平均显著降低(P<0.05,P<0.01)。结论:电针可能通过介导FOXO Objective To explore the effects of electroacupuncture(EA) on skeletal muscle and blood glucose in rats with diabetic amyotrophy. Methods Among 40 SD rats, 10 rats were randomly selected into the control group and received no treatment. The remaining 30 rats were treated with intraperitoneal injection of streptozotocin(STZ, 60 mg/kg) to establish diabetes mellitus(DM) model, and then the rats were treated with vascular ligation at right posterior limb to establish amyotrophy model. The rats with diabetic amyotrophy were randomly divided into a model group and an EA group, 10 rats in each group(10 rats were excluded due to unsuccessful model establishment and death). The rats in the EA group was treated with EA at right-side "Yishu(EX-B 3)" "Shenshu(BL 23)" "Zusanli(ST 36)" and "Sanyinjiao(SP 6)", disperse-dense wave, 2 Hz/15 Hz, 20 minutes each time, once a day for 3 weeks. Before and after EA treatment, the blood sample was collected from inner canthus and the "glucose oxidase-peroxidase" method was used to detect fasting blood glucose level;ELISA method was used to detect insulin content. At the end of the treatment, HE staining method was used to observe the morphology of ischemic skeletal muscle in the right hindlimb;the real-time PCR method was used to detect the m RNA expression of muscle atrophy F-box(MAFbx), muscle ring finger-1(Mu RF1) and forkhead box O3 a(FOXO3 a) in the ischemic skeletal muscle tissue of right hindlimb. Results Before the treatment, the body mass in the model group and EA group was lower than that in the control group(P<0.01);after the treatment, the body mass in the control group was increased, while the body mass in the model group and EA group was decreased(P<0.01). Compared with the control group, the fasting blood glucose was significantly increased and insulin content was significantly decreased in the model group(P<0.01);compared with the model group, the fasting blood glucose was significantly decreased and the insulin content was significantly increased in the EA group after
