基于油水分配系数与指标成分含量的甘草增溶作用研究 被引量:3

摘要 目的:通过测定桃核承气汤中除芒硝外各味药单煎液、单味药分别加甘草的合煎液、全方药不加甘草的合煎液及全方药物合煎液的油水分配系数(logP)与指标成分的含量,验证甘草增溶原理。方法:用正辛醇萃取各煎液分离其油相与水相,水浴浓缩后称重,计算得到各煎液油水分配系数;采用高效液相色谱法(HPLC)测定单煎液与合煎液中苦杏仁苷、桂皮醛、甘草酸、大黄酸、大黄素、大黄酚、大黄素甲醚及芦荟大黄素含量。结果:桃核承气汤中单味药材加甘草后油水分配系数均减少0.4547及以上;指标成分含量均有所增加。结论:通过对桃核承气汤中油水分配系数与指标成分含量的研究,验证了甘草的增溶作用,并进一步说明甘草增溶通过甘草酸形成胶束来实现,为甘草在临床应用中可增加其他药物有效成分在水中的溶解度,提高制剂中有效成分含量,增强临床疗效提供理论基础。 Objective:To determine the content of oil-water distribution coefficient(logP)and the content determination of indicator components of Taohe Chengqi Decoction except mirabilite,the combination decoction of licorice,the whole prescription decoction without licorice and the whole prescription decoction,so as to verify the solubilization effect of licorice.Methods:The oil phase and water phase were separated by n-octanol extraction and weighed after concentration in water bath,and the oil-water partition coefficients of each decoction were calculated,and the contents of bitter almond glycoside,cinnamaldehyde,glycyrrhizic acid,emodin,chrysophanol,emodin methyl ether and aloe emodin in single decoction and combined decoction were determined by high performance liquid chromatography(HPLC).Results:The results showed that oil-water distribution coefficient of Taohe Chengqi decoction decreased by 0.4547 or more,and the content of index components increased after adding licorice in Taohe Chengqi Decoction.Conclusion:The research on oil-water partition coefficient and the content of the index components in Taohe Chengqi Decoction verified the solubilization effect of licorice,and further explained that the solubilization of licorice was achieved by the formation of micelles of glycyrrhizic acid,which could be used to increase the solubility of the active ingredients in water,which can increase the content of the active ingredients in the preparation and provide a theoretical basis for enhancing the clinical efficacy.
