配伍对十味温胆汤中丹参特征图谱的比较研究 被引量:2

摘要 目的建立十味温胆汤中丹参的特征图谱,研究配伍对十味温胆汤中丹参特征峰的影响。方法应用HPLC法,色谱柱为Kromasil-C18(4.6 mm×250 mm,5μm),乙腈(A)^-1.0%冰乙酸水(B)为流动相,梯度洗脱,体积流量为1.0 mL·min^-1,检测波长254 nm。结果以十味温胆汤为参比峰,标定了十味温胆汤中的14个特征峰,其中1、5、7、9、10号峰是丹参与十味温胆汤的共有峰,5、7、13、14号是十味温胆汤去丹参的色谱峰。结论与单味丹参相比,全方中丹参特征峰峰面积均有不同程度的降低,配伍对十味温胆汤中丹参特征峰有较为显著的影响,使丹参中的某些成分溶出量减少,这可能与配伍后丹参与十味温胆汤各药某些成分结合使其溶出减少所致,也可能与十味温胆汤煎煮时间、丹参剂量有关联。 Objective To establish the specific chromatogram of Danshen in Shiwei Wendan decoction,and to study the effect of combination on the characteristic peak of Danshen in Shiwei Wendan decoction.Methods kromasil-c18(4.6 mm×250 mm,5μm)chromatographic column,acetonitrile(A)^-1.0%glacial acetic acid(B)as mobile phase,with gradient elution,flow rate of 1.0 ml·min^-1,detection wavelength of 254 nm,column temperature of 35℃.Results With Shiwei Wendan decoction as the reference peak,14 characteristic peaks in Shiwei Wendan decoction were demarcated.Among them peaks 1,5,7,9 and 10 are the common peaks of Danshen and Shiwei Wendan decoction.Peaks 5,7,13 and 14 are the chromatographic peaks of Shiwei Wendan Decoction without Danshen.Conclusion compared to decoction with only Danshen,the peak area of the whole Shiwei Wendan decoction all are reduced with varying degrees,it means combination has a significant effect on the Danshen’s characteristics peak in Shiwei Wendan decoction,dissolution of some Danshen ingredients reduced because of combination.It was speculated that Danshen dissolved less when it binds with some other components of Shiwei Wendan decoction after combination,and may also be related to the decocting time or the dosage of Danshen in Shiwei Wendan decoction.
