摘要
目的:探讨低浓度苯和甲醛经呼吸道联合染毒对雌性ICR小鼠的肝脏毒性,为职业场所苯和甲醛联合暴露导致的健康效应提供实验参考和科学依据。方法:54只SPF级雌性ICR小鼠随机分组均分9组,依次分为G 1(空白对照组)、G 2(低苯组,苯剂量12 mg·m^-3)、G 3(高苯组,苯剂量60 mg·m^-3)、G 4(低甲醛组,甲醛剂量2 mg·m^-3)、G 5(高甲醛组,甲醛剂量10 mg·m^-3)、G 6(低苯+低甲醛组,苯剂量12 mg·m^-3+甲醛剂量2 mg·m^-3)、G 7(高苯+低甲醛组,苯剂量60 mg·m^-3+甲醛剂量2 mg·m^-3)、G 8(低苯+高甲醛组,苯剂量12 mg·m^-3+甲醛剂量10 mg·m^-3)、G 9(高苯+高甲醛组,苯剂量60 mg·m^-3+甲醛剂量10 mg·m^-3),每组6只,呼吸道静式染毒,连续染毒14天,2小时/天(中间间隔1小时)。染毒期间观察小鼠的一般情况并每周称量体重,染毒结束前12小时禁食禁水称量终点体重,染毒结束动物麻醉后采血分离血清测定丙氨酸转氨酶(ALT)和天门冬氨酸转氨酶(AST)、血清总胆固醇(CHO)、血清甘油三脂(TG),并制备10%肝匀浆测定肝脏组织超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量。结果:①苯不同剂量组小鼠的体重变化不同,差异有统计学意义(P<0.05),苯和甲醛对体重的增长有交互作用,差异有统计学意义(P<0.05);②苯和甲醛对AST有交互作用,差异有统计学意义(P<0.05);③苯不同剂量组MDA的变化不同,差异有统计学意义(P<0.05),甲醛不同剂量组MDA的变化不同,差异有统计学意义(P<0.05),苯和甲醛对MAD的变化有交互作用,差异有统计学意义(P<0.05);④苯不同剂量组小鼠的TG不同,差异有统计学意义(P<0.05),苯和甲醛对TG有交互作用,差异有统计学意义(P<0.05)。结论:①低浓度、短时间的苯和甲醛单独暴露对ICR小鼠具有系统毒性及肝脏毒性,与甲醛相比,苯的肝脏毒效应更显著;②低浓度、短时间的苯和甲醛联合暴露对ICR小鼠导致的系统毒性及肝脏毒性有交互作用,
Objective:To explore the liver toxicity of low concentration of benzene(BZ)and formaldehyde(FA)joint exposure to the female ICR mice through static respiratory tract,to provide experimental reference and scientific foundation for prevention and control of joint exposure of benzene and formaldehyde in occupational places.Methods:54 SPF female ICR mice were divided randomly into 9 groups,including G 1(control group),G 2(low BZ group,BZ dose 12 mg·m^-3),G 3(high BZ group,BZ dose 60 mg·m^-3),G 4(low FA group,FA dose 2 mg·m^-3),G 5(high FA group,FA dose 10 mg·m^-3),G 6(low BZ+low FA group,BZ dose 12 mg·m^-3+FA dose 2 mg·m^-3),G 7(high BZ+low FA group,BZ dose 60 mg·m^-3+FA dose 2 mg·m^-3),G 8(low BZ+high FA group,BZ dose 12 mg·m^-3+FA dose 10 mg·m^-3),G 9(high BZ+high FA group,BZ dose 60 mg·m^-3+FA dose 10 mg·m^-3),6 in each group,the mice of G1 group were treated with air as the blank control group,all mice were exposed with static respiratory tract,continuous exposure for 14 days,2 hours/day(interval of one hour).The general condition of the mice was observed during the exposure period and weight was measured weekly.Ban food and water during 12 hours before experiment terminal and measure the last weight,mice were anaesthetized and collected serum for testing AST,ALT,CHO and TG,10%liver homogenates were made for testing SOD and MDA.Results:①The difference in weight was significant in mice between different groups of benzene(P<0.05),there was remarkable interaction for B and FA in weight(P<0.05);②There was remarkable interaction for B and FA in ALT(P<0.05);③The difference in MDA was significant in mice between different groups of benzene(P<0.05),the difference in MDA was significant in mice between different groups of formaldehyde(P<0.05),there was remarkable interaction for B and FA in MDA(P<0.05);④The difference in TG was significant in mice between different groups of benzene(P<0.05),there was remarkable interaction for B and FA in TG(P<0.05).Conclusions:①Low concentration,short exposure of be
