基于花粉形态与叶片形态的金柑属种质资源亲缘关系分析 被引量:5

Genetic Relationship Analysis of Fortunella Germplasm Resources Based on Pollen and Leaf Morphology

摘要 本研究利用扫描电镜(SEM)对金柑属、柑橘属和枳属36个不同品种的花粉形态特征进行了观察研究,并结合叶形态观察结果进行了比较分析。结果表明,金柑属花粉粒大小为(21.18~31.35)μm×(14.05~26.18)μm,网脊宽为0.52~0.98μm,网眼为圆形或近圆形,直径大小为0.48~1.62μm;柑橘属花粉粒大小为(25.99~32.53)μm×(17.54~20.05)μm,网脊宽为0.72~0.92μm,网眼直径大小为0.51~1.10μm,多为圆形或近圆形;枳属花粉粒大小为(31.03~32.60)μm×(18.82~19.36)μm,网脊宽为0.38~0.40μm,网眼多为不规则多角形,网眼直径大小为1.45~1.46μm。金柑属和柑橘属的花粉粒大多数都具有4条萌发沟,2份枳属材料具有5条萌发沟。‘富圆金柑’具有较复杂的浮雕状纹饰,其余品种的花粉粒均为网状或细网状纹饰。花粉聚类分析结果显示当欧氏距离为10时,可将36份材料分为4个分支。叶的基本形态具有属的特异性,与花粉聚类分析推断出的亲缘关系一致。本研究结果可以为金柑不同种属的亲缘关系以及品种鉴定提供依据。 The morphological characteristics of pollen were observed by scanning electron microscopy(SEM)in 36 different varieties of Fortunella,Citrus and Poncirus.Their leaf morphology observation were compared and analyzed.The pollen grains(P×E)of Fortunella Swing were(21.18~31.35)μm×(14.05~26.18)μm;The widths of mesh ridges were 0.52~0.98μm;The mesh was round or nearly-round,and the mesh sizes were 0.48~1.62μm;The pollen grains(P×E)of Citrus were(25.99~32.53)μm×(17.54~20.05)μm;The widths of mesh ridges were 0.72~0.92μm;And the mesh sizes were 0.51~1.10μm,mostly round or nearly-round;The pollen grains(P×E)of the Poncirus were(31.03~32.60)μm×(18.82~19.36)μm;The widths of mesh ridge were 0.38~0.40μm;The meshes were mostly irregular polygon,the mesh sizes were 1.45~1.46μm.The pollens of most varieties were 4-colpate type.Two Poncirus varieties were 5-colpate type.The surface structures of Fortunella pollen grains are mesh or fine mesh except F.crassifolia Swing.'Fuyuanjingan'with complex exine sculpture.The clustering analysis based on pollen morphology indicated that 36 varieties could be classified into 4 groups at the euclidean distance of 10.The morphology of the leaves is specific among genus,which is consistent with the genetic relationship inferred from the pollen clustering analysis.The study can provide a basis for the genetic relationship and cultivar identification of different kumquat species.
