脑栓通胶囊对急性脑梗死模型大鼠炎症因子、MMP-9和TIMP-1表达及氧化应激指标的影响 被引量:10

摘要 目的探讨脑栓通胶囊对急性脑梗死模型大鼠炎症因子、MMP-9和TIMP-1表达及氧化应激指标的影响。方法将60只SD大鼠随机分为假手术组、模型组、阳性组和实验组,采用改良线栓法制作急性脑梗死大鼠模型。假手术组自由饮水进食,模型组灌服纯净水,实验组灌服脑栓通胶囊,剂量120 mg/kg,阳性组以10 mg/kg灌服尼莫地平,每日1次,连续7 d。7 d后,测评各组大鼠的神经功能评分,观察比较各组大鼠炎症因子、MMP-9和TIMP-1表达及氧化应激指标。结果与假手术组比较,模型组大鼠的神经功能评分和神经行为学评分、TNF-α、CRP、IL-1β以及IL-6水平、MMP-9基因表达和MMP-9蛋白表达的相对丰度值以及MDA水平明显升高,TIMP-1基因表达和TIMP-1蛋白表达的相对丰度值、SOD、GSH以及CAT水平明显降低,具有显著差异(P<0.05)。给予药物干预后,阳性组和实验组大鼠的神经功能评分和神经行为学评分、炎症因子、MMP-9基因表达和MMP-9蛋白表达明显低于模型组(P<0.05),TIMP-1基因表达和TIMP-1蛋白表达的相对丰度值、SOD、GSH以及CAT水平明显高于模型组(P<0.05)。结论脑栓通胶囊可有效降低急性脑梗死模型大鼠炎症因子水平,改善MMP-9和TIMP-1表达,降低氧化应激指标。 Objective:To investigate the effects of Naoshuantong Capsule on the expression of inflammatory factors,MMP-9 and TIMP-1 and oxidative stress in rats with acute cerebral infarction.Methods:60 SD rats were randomly divided into sham operation group,the model group,the positive group,and the test group.The rat model of acute cerebral infarction was made with a modified suture method.The sham operation group drank water freely.The model group began to irrigate pure water.The experimental group was given Naoshuantong Capsule with a dose of 120 mg/kg.The positive group received nimodipine with a dose of 10 mg/kg,once a day,continuously for 7 d.After 7 days,the neurological scores were evaluated,and the expressions of inflammatory factors,MMP-9 and TIMP-1 and oxidative stress indicators were observed and compared.Results:Compared with the sham operated group,the neural function score,neurobehavioral score,the levels of TNF-α,CRP,IL-1βand IL-6,the relative abundance value of MMP-9 gene expression and protein expression and MDA level were significantly higher;the relative abundance value of TIMP-1 gene expression and protein expression and the levels of SOD,GSH and CAT decreased significantly in the model group(P<0.05).After drug intervention,compared with the model group,the neurological function score,neurobehavioral score,inflammatory factors,MMP-9 gene expression and protein expression were significantly lower,and the relative abundance value of TIMP-1 gene expression and protein expression and the levels of SOD,GSH and CAT were higher in the positive group and the experimental group(P<0.05).Conclusion:Naoshuantong Capsule can effectively reduce the levels of inflammatory factors,improve the expression of MMP-9 and TIMP-1,and reduce the index of oxidative stress in rats with acute cerebral infarction.
