摘要
目的探讨胡桃醌对人卵巢癌细胞上皮间质转化的作用及机制。方法胡桃醌作用SKOV3细胞24 h,细胞划痕实验检测细胞迁移能力;免疫印迹实验检测E-钙黏蛋白、N-钙黏蛋白、NF-κB p65和Snail蛋白表达水平;使用NF-κB途径抑制剂PDTC和胡桃醌联合作用SKOV3细胞后,免疫印迹实验检测Snail蛋白表达。结果细胞划痕实验结果表明,胡桃醌作用SKOV3细胞后迁移能力下降。免疫印迹实验结果表明,胡桃醌作用SKOV3细胞后E-钙黏蛋白表达升高,N-钙黏蛋白、Snail和NF-κB p65蛋白表达下降。与胡桃醌组比较,胡桃醌与PDTC联合作用SKOV3细胞后Snail蛋白表达下降。结论胡桃醌可能通过抑制NF-κB/Snail信号通路,从而抑制SKOV3细胞上皮间质转化。
Objective To investigate the effect and mechanism of juglone on epithelial mesenchymal transformation of human ovarian cancer cells.Methods The SKOV3 cells were treated with juglone for 24 hours,and the migration ability was detected by scratch test.The expression levels of E-cadherin,N-cadherin,NF-κB p65 and Snail were detected by immunoblotting.The expression of Snail was detected by immunoblotting after SKOV3 cells were treated with PDTC,an inhibitor of NF-κB pathway,and juglone.Results The results of cell scratch test showed that the migration ability of SKOV3 cells decreased after juglone treatment.The results of Western blotting showed that the expression of E-cadherin increased,while the expression of N-cadherin,Snail and NF-κB p65 decreased.Compared with the walnut quinone group,the expression of Snail protein in SKOV3 cells decreased after the combination of walnut quinone and PDTC.Conclusion It is suggested that juglone may inhibit the epithelial stromal transformation of SKOV3 cells by inhibiting NF-κB/Snail signaling pathway.
