化痰散结方含药血清作用胃癌细胞系MGC803后差异表达miRNAs筛选及生物信息学分析 被引量:1

摘要 目的:筛选化痰散结方含药血清处理胃癌细胞系MGC803后差异表达的微小RNA(miRNAs),并进行生物信息学分析。方法:应用"下一代"测序技术(即高通量测序技术)对MGC803细胞和化痰散结方含药血清处理后的MGC803细胞进行miRNAs差异表达谱分析;结合文献报道确定4个与肿瘤相关的miRNAs,对其进行靶基因预测及信号转导通路的分析。结果:经化痰散结方含药血清处理后,miR-424~3p和miR-494~3p的表达水平明显上调,miR-33b-3p和miR-27a-5p的表达水平明显下调;生物信息学方法分析结果显示miR-424~3p、miR-494~3p、miR-27a-5p和miR-33b-3p及其靶基因参与细胞迁移、细胞周期和凋亡等通路。结论:化痰散结方含药血清可能通过调控miR-27a-5p、miR-33b-3p、miR-424~3p、和miR-494~3p的表达抑制胃癌的发生发展。 Objective: Screening differentially expressed microRNAs(miRNAs) after treatment of gastric cancer cell line MGC803 with Huatan Sanjie Prescription containing serum,and conducting bioinformatics analysis.Methods: High-throughput sequencing technology was used to analyze the differential expression profiles of miRNAs in MGC803 cells and Huatan Sanjie Prescription containing serum treated MGC803 cells.Four tumor-associated miRNAs were identified in the literature,and target gene prediction and signal transduction pathways were analyzed.Results: After treatment with Huatan Sanjie Prescription containing serum,the expression levels of miR-424~3 p and miR-494~3 p were significantly up-regulated,and the expression level of miR-27 a-5 p and miR-33 b-3 p was significantly down-regulated.Bioinformatics analysis showed that miR-424~3 p,miR-494~3 p,miR-27 a-5 p and miR-33 b-3 p and their target genes were involved in cell migration,cell cycle and apoptosise etc.Conclusion: The Huatan Sanjie Prescription containing serum may participate in the development of gastric cancer through the expression of miR-33 b-3 p,miR-27 a-5 p,miR-424~3 p and miR-494~3 p.
