摘要
目的探讨真核延伸因子-1A2(eEF1A2)基因对宫颈癌细胞增殖、侵袭和迁移的影响。方法设计eEF1A2基因的siRNA干扰片段分别转染宫颈癌SiHa细胞、HeLa细胞和C33A细胞,实验组包括:转染Si-eEF1A2-1和SieEF1A2-2的细胞组,未转染的细胞和非干扰转染细胞(NC组)为对照组。采用Western blot方法检测eEF1A2和E-cadherin在宫颈癌细胞中蛋白质表达水平,通过划痕实验和基质胶包被transwell实验分析细胞迁移能力,通过CCK-8实验和平板克隆实验检测细胞增殖。结果宫颈癌细胞中,siRNA干扰的细胞与对照组相比eEF1A2蛋白质表达水平降低,E-cadherin蛋白质表达水平升高,差异具有统计学意义(P<0.05);划痕实验si-eEF1A2转染的细胞与对照组相比较,愈合间隙宽,差异有统计学意义(P<0.05);Transwell迁移实验,RNA干扰的细胞与对照组相比,穿过小室基底膜的细胞明显少于对照组,差异有统计学意义(P<0.05);si-eEF1A2干扰的细胞CCK8增殖实验、细胞克隆形成实验中可以抑制宫颈癌细胞增殖,差异有统计学意义(P<0.05)。结论降低宫颈癌SiHa、HeLa和C33A细胞eEF1A2基因表达水平,可以抑制癌细胞的侵袭和迁移。
Objective To investigate the effect of eukaryotic elongation factor-1A2(eEF1A2)gene on the proliferation,invasion and migration of cervical cancer cells.Methods The siRNA interference fragments of the eEF1A2 gene were designed to transfect cervical cancer cell SiHa,HeLa and C33A respectively.The experimental group included:Cell groups transfected with Si-eEF1A2-1 and Si-eEF1A2-2,untransfected cells and non-interfering transfected cells(NC group)as control group.Western blot was used to detect the protein expression level of eEF1A2 and E-cadherin in cervical cancer tissue.Cell migration ability was analyzed by scratch test and matrigel-coated transwell test.Cell proliferation was measured by CCK-8 test and plate cloning test.Results In cervical cancer cells,compared with the control group,the protein expression level of siRNA interference cells decreased,and the expression level of E-cadherin protein increased.The difference was statistically significant(P<0.05).Cells transfected with si-eEF1A2 in the scratch test compared with the control group,the healing gap was wide,and the difference was statistically significant(P<0.05).In the Transwell migration test,compared with the control group,the cells that passed through the cell basement membrane were significantly less than the control group.The difference was statistically significant(P<0.05).Si-eEF1A2 interfered with cell CCK8 proliferation experiments and cell clone formation experiments could inhibit cervical cancer cell proliferation,and the differences were statistically significant(P<0.05).Conclusion Decreasing the expression level of eEF1A2 gene in cervical cancer SiHa,HeLa and C33A cells can inhibit the invasion and migration of cancer cells.
