摘要
目的检测广西壮族绝经妇女雌激素受体(ER)-α基因分布,并检测不同ER-α基因型对应血清女性性激素水平,明确不同ER-α基因的绝经后妇女其卵巢功能退化情况。方法对45~50岁的壮族妇女(376例)采集外周5 ml静脉血,用聚合酶链式反应-限制性片段长度多态分析技术检测,使用PvuⅡ酶和XbaⅠ酶进行酶切,得出不同ER-α基因型,对结果统计分析。用放免发光法检测各基因型ER-α基因型样本的血清各种女性激素含量。结果5个ER-α的PvuⅡ酶切基因(PP,Pp,pp,p,P)的血清女性激素水平中,PP基因型,Pp基因型,pp基因型,p基因型之间相比较,4基因型之间两两比较,差异无显著性(P>0.05)。与其他4组比较,P基因型的血清雌激素和孕激素明显偏低(P<0.05),促卵泡生产素和促黄体素明显偏高(P<0.01)。5个ER-α的XbaⅠ酶切得到基因型(XX,Xx,xx,x,X)组的血清雌激素、孕激素水平中,XX基因型,xx基因型,x基因型和X基因型之间相比较,4个基因型之间差异无显著性(P>0.05)。与其他4个基因型比较,Xx基因型的雌激素和孕激素血液水平明显增高(P<0.05),血液促卵泡生长素、促黄体明显偏低(P<0.01)。结论ER-αPvuⅡ酶切的P等位基因型的女性卵巢功能退化更早,雌激素和孕激素血液水平下降更为明显,应提前做好老年女性激素退化所致一系列疾病的防治。而ER-αXbaⅠ的Xx等位基因型壮族妇女,卵巢功能退化较晚,不易患雌激素和孕激素缺乏所致老年性疾病。
Objective To detect estrogen receptor(ER)-αgenotype and corresponding serum women’s sex hormone level in Zhuang women after menopause.Methods A total of 376 Zhuang(Guangxi)45~50 years old menopausal women were routinely administrated with 5 ml of venous blood in the morning,and the distribution of ER-αgene was detected and analyzed by PCR-RFLP.The serum samples of each genotype were determined by radioimmunoassay for follicle stimulating hormone,luteinizing hormone and estradiol and progesterone.Results Among PP,Pp,pp,p,P genome(ER-αgene PvuⅡrestriction gene),there were no significant differences in serum sex hormones among PP,Pp,pp,p genomes groups(P>0.05),while there were significant differences in serum sex hormones between P genome and the other four groups(P<0.05):the blood levels of estradiol and progesterone were significantly lower than those of the other 4 groups(P<0.05);the follicle stimulating hormone and luteinizing hormone were significantly higher than the other 4 groups(P<0.01)in P genome group.Among XX,Xx,xx,x,X genome(ER-αgene Xba I restriction gene),there were no significant differences in serum estradiol and progesterone among XX,xx,x,X genomes groups(P>0.05),there were significant differences in serum estradiol and progesterone between Xx genome and the other 4 groups(P<0.01):the estradiol and progesterone levels in the Xx genome were significantly higher than those of the other groups(P<0.05);while follicle stimulating hormone and luteinizing hormone in Xx genome were significantly lower than those of the other 4 groups(P<0.01).Conclusions The ER-αPvuⅡdigested P allele in menopausal women has a lower level of estrogen and progesterone are more prone to postmenopausal estrogen deficiency senile diseases.The Xx allelic menopausal women of ER-αXbaⅠhave higher levels of estrogen and progesterone and are less prone to estrogen deficiency.
