生血补髓方对激素性股骨头缺血性坏死模型大鼠血清OPG、RANK、RANKL含量的影响 被引量:10

Effects of Shengxue Busui Fang( 生血补髓方) on serum level of OPG,RANK and RANKL in rats model with steroid-induced avascular necrosis of femoral head( SANFH)

摘要 目的通过研究生血补髓方对激素性股骨头缺血性坏死模型大鼠血清护骨素(OPG)、核因子κB受体活化因子(RANK)及核因子κB受体活化因子配体(RANKL)含量的影响,探讨生血补髓方预防激素性股骨头缺血性坏死的作用机制。方法采用随机数字表法将40只SD大鼠分为空白组、模型组、福善美组、生血补髓方组4组,每组10只。除空白组外,其余各组大鼠通过注射大肠埃希杆菌内毒素和强的松龙建立激素性股骨头缺血性坏死模型。造模成功后,各组灌胃生理盐水及相应药物,灌胃8周后采用双抗夹心酶联免疫吸附测定(ELISA)法检测各组大鼠血清OPG、RANK、RANKL的含量。结果与空白组比较,模型组大鼠血清RANK、RANKL含量明显升高,OPG含量明显降低,差异均有统计学意义(P<0.01);与模型组比较,福善美组、生血补髓方组大鼠血清RANK、RANKL含量明显降低,OPG含量明显升高,差异均有统计学意义(P<0.01)。结论生血补髓方能调节激素性股骨头缺血性坏死模型大鼠血清OPG、RANK、RANKL的表达,抑制破骨细胞的活化,促进新骨的形成及骨修复,使骨代谢恢复平衡。 Objective To discuss the action mechanism of Shengxue Busui Fang( 生血补髓方,SBF) in prevention of steroid-induced avascular necrosis of femoral head( SANFH),by investigating the effect thereof on serum level osteoprotegerin( OPG),receptor activator nuclear factor-κB( RANK) and receptor activator nuclear factor-κB ligand( RANKL) in rats model with SANFH. Methods Forty SD rats were divided into 4 groups including blank group,model group,Fushanmei group and SBF group,each with 10. Except the blank group,the rats in the other groups were given injection of Escherichia coli endotoxin and prednisolone to establish SANFH models. After the successful establishment of the models,each group was given normal saline and the corresponding drugs by gavage.After 8 weeks of administration by gavage,the double-antibody sandwich enzyme linked immunosorbent assay( ELISA) method was used to determine the serum level of OPG,RANK and RANKL in rats of each group.Results Compared with the blank group,the serum level of RANK and RANKL in rats of the model group significantly increased,the level of OPG significantly decreased,and the difference was statistically significant( P < 0. 01). Compared with the model group,the serum level of RANK and RANKL in rats of Fushanmei group and SBF group significantly decreased,while the level of OPG significantly increased,and the difference was statistically significant( P<0.01).Conclusion SBF can regulate the serum level of OPG,RANK and RANKL in rats model with SANFH,inhibit the activation of osteoclasts,promote the formation of new bone and bone repair,and restore balance to bone metabolism.
