桥联双β-环糊精液相色谱柱测定药片中盐酸阿罗洛尔对映体

Determination of arotinolol hydrochloride enantiomers in tablets on a bridged bis(β-cyclodextrin)column in HPLC

摘要 采用一种自制的桥联β-环糊精手性色谱柱(UBCDP),通过流动相组成、流速、柱温等色谱条件的优化,在极性有机的模式下成功地拆分了盐酸阿罗洛尔对映体,分离度在25 min内可达到1.58。在上述基础上,建立了一种高效液相色谱法荧光法测定药片中盐酸阿罗洛尔对映体含量的新方法,在0.05~0.50 mg·L-1的浓度范围内两个对映体均具有良好的线性关系(R2≥0.9992),平均回收率范围为92.8~105.1%(n=5),检出限(LOD)低于0.025 mg·L-1。该方法操作较简便,选择性较好,灵敏度高,分析时间较短,为盐酸阿罗洛尔手性药物的质量控制和相关药代动力学研究提供了一种新方法。 A home-made bridgedβ-cyclodextrin chiral chromatographic column(UBCDP)was successfully used to separate the enantiomers of arotinol hydrochloride in the polar organic mode,and the chromatographic conditions such as mobile phase composition,flow rate and column temperature were optimized.The resolution could arrive at 1.58 within 25 min.Furthermore,a new method for determination of arotinolol hydrochloride enantiomers in tablets by high performance liquid chromatography with fluorescence detector has been established.In this study,it could be found that the two enantiomers had good linear relationship(R2≥0.9992)in the concentration range of 0.05~0.50 mg·L-1,also the average recovery range was 92.8~105.1%(n=5),and the detection limit(LOD)was less than 0.025 mg·L-1.The method is simple and convenient to operate.Moreover,it had high selectivity and sensitivity,which provided a new method for quality control and pharmacokinetic study of arotinolol hydrochloride enantiomers.
