基于高通量数据分析DNA甲基化和拷贝数畸变对C1orf26基因表达的影响

Analysis of DNA methylation and copy number aberrations on C1orf26 gene expression in breast cancer based on high-throughput multiomics databases

摘要 目的:揭示乳腺癌中1号染色体开放阅读框26(Chromosome 1 Open Reading Frame 26,C1orf26)DNA甲基化和拷贝数畸变对表达的影响。方法:利用癌症多组学和临床数据库LinkedOmics分析DNA甲基化和拷贝数畸变对乳腺癌组织中C1orf26基因表达的影响。结果:C1orf26 mRNA水平与其基因拷贝数变化拷贝数畸变正相关(P<0.01),而与其DNA甲基化水平负相关(P<0.01);且LinkedOmics分析结果显示乳腺癌中C1orf26 mRNA水平分别受到组织学类型、肿瘤纯度和患者种族影响(P<0.05、P<0.01、P<0.01)。结论:C1orf26基因拷贝数畸变和DNA甲基化水平影响其表达,C1orf26可作为乳腺癌预后的潜在标志物。 Objective To reveal the effect of DNA methylation and copy number aberrations on expression of Chromosome 1 Open Reading Frame 26(C1orf26)in breast cancer.Method the cancer multi-omics and clinical database LinkedOmics were used to analyze changes of C1orf26 gene in breast cancer.Results Results from LinkedOmics showed that C1orf26 mRNA level was positively correlated with its gene copy-number alteration(P<0.01),and was negatively correlated with its DNA methylation(P<0.01).C1orf26 expression was significantly affected by histological type,Tumor purity and patient ethnicity(P<0.05,P<0.01,P<0.01).Conclusion Gene copy number alteration and DNA methylation promote C1orf26 expression,and C1orf26 could become a candidate prognostic marker for breast cancers.
