摘要
目的:探讨丹皮酚对表柔比星诱导膀胱癌BIU-87细胞凋亡的影响及机制。方法:实验分为BIU-87细胞组、表柔比星组(100μg·ml-1)、丹皮酚组(100μg·ml-1)、表柔比星+丹皮酚组(100μg·ml-1+100μg·ml-1),各组细胞分别设6个平行样,培养72 h。培养结束后应用CCK-8试剂盒测定细胞增殖水平,吉姆萨染色测定菌落百分比,膜联蛋白V-碘化丙啶凋亡检测试剂盒分析细胞凋亡水平,流式细胞仪测定细胞周期、逆转录-聚合酶链反应(RT-PCR)及免疫印迹试验(Western blot)测定BIU-87细胞B淋巴细胞瘤-W(Bcl-w)、半胱氨酸蛋白酶-6(Caspase-6)基因及蛋白水平。结果:与BIU-87细胞组比较,表柔比星组、丹皮酚组、表柔比星+丹皮酚组的细胞存活率、细胞菌落百分比明显降低(P<0.05),细胞凋亡率、G1期、Bcl-w、Caspase-6 mRNA及蛋白表达水平明显升高(P<0.05)。与表柔比星组、丹皮酚组比较,表柔比星+丹皮酚组的细胞存活率、细胞菌落百分比明显降低(P<0.05),细胞凋亡率、G1期、Bcl-w、Caspase-6 mRNA及蛋白表达水平明显升高(P<0.05)。结论:丹皮酚对表柔比星抑制膀胱癌BIU-87增殖、诱导膀胱癌细胞凋亡具有叠加作用;其机制与表柔比星、丹皮酚能促进膀胱癌BIU-87细胞Bcl-w和Caspase-6 mRNA及蛋白的表达有关。表柔比星联合丹皮酚使用时,其对Bcl-w、Caspase-6 mRNA和蛋白表达的促进作用尤为明显。
Objective:To investigate the effect of paeonol on epirubicin-induced apoptosis of bladder cancer BIU-87 cells and its mechanisms.Methods:BIU-87 cell group,epirubicin group(100μg·ml-1),paeonol group(100μg·ml-1)and epirubicin+paeonol group(100μg·ml-1+100μg·ml-1)were set up.The cells in each group were divided into 6 parallel samples for 72 h.CCK-8 kit was used to detect the cell proliferation level,Jimsa staining was used to determine the percentage of colonies,annexin V-propidium iodide apoptosis detection kit was used to analyze the apoptotic level,flow cytometry was used to determine the cell cycle,and RT-PCR and Western blot were used to determine Bcl-w,Caspase-6 mRNA and protein levels in BIU-87 cells.Results:Compared with those in BIU-87 cell group,the survival rate and percentage of cell colonies were significantly lower in epirubicin group,paeonol group and epirubicin+paeonol group(P<0.05),and the apoptotic rate,G1 phase,Bcl-w,Caspase-6 mRNA and protein levels were significantly increased(P<0.05).Compared with those in epirubicin group and paeonol group,the survival rate and percentage of cell colonies were significantly lower in epirubicin+paeonol group(P<0.05),and the apoptotic rate,G1 phase,Bcl-w,Caspase-6 mRNA and protein levels were significantly increased(P<0.05).Conclusion:Paeonol can inhibit the proliferation and induce the apoptosis of bladder cancer cells.The mechanism is related to the promotion of Bcl-w,Caspase-6 mRNA and protein expressions of bladder cancer BIU-87 cells by epirubicin and paeonol.Epirubicin combined with paeonol can especially promote the expressions of Bcl-w,Caspase-6 mRNA and protein.
