摘要
梅花鹿鹿茸是一种传统名贵中药材,在我国已沿用两千多年。本研究通过LC-MS-MS技术对梅花鹿鹿茸的脂质组分进行比较分析。结果表明,共获得752种梅花鹿鹿茸脂质成分,共分为15个类别,其中磷脂酰乙醇胺相对含量最高,其次为磷脂酰胆碱和甘油三酯;利用OPLS-DA多元统计方法与单因素方差检验筛选到133种显著差异表达脂质分子(VIP> 1,P<0.05,差异倍数> 2),包含58种甘油三脂,22种磷脂酰胆碱,7种磷脂酰丝氨酸,35种磷脂酰乙醇胺,2种溶血磷脂酰胆碱,5种溶血磷脂酰乙醇胺和4种神经酰胺;相对定量结果显示,甘油三脂与磷脂类化合物在蜡片茸中高表达,而能够维持皮肤水分、调节细胞凋亡的神经酰胺类化合物在血片茸中高表达;另外,筛选到5种甘油三脂在三杈蜡片茸相对含量显著高于二杠蜡片茸。本研究详细分析了蜡片茸与血片茸的脂质组分,为鹿茸有效活性成分的挖掘与鹿茸生长机制的探究奠定了理论基础。
Antler is a kind of valuable traditional Chinese medicine,which has been used in China for more than two thousand years.The purpose of this study was to compare the lipid components of antler in sika deer by LC-MS-MS.In this study,a total of 752 lipid components of antler in sika deer were obtained,which were divided into 15 categories,among which the relative content of phosphoethanolamine was the highest,followed by phosphatidylcholine and triglyceride.133 significant difference expression of lipid molecules(VIP>1,P<0.05,differences in multiples>2)were screened by OPLS-DA multivariate statistical method and single factor variance test,containing 58 trig-lycerides,22 phosphatidylcholines,7 phosphatidylserines,35 phosphatidylethanolamines,2 lysophosphatidylcholine,5 lysophosphatidylthanolamines and 4 ceramides.The relative quantitative results showed that triglycerides and phospholipids were highly expressed in ant-ler wax slices,while ceramides,which could maintain skin moisture and regulate cell apoptosis,were highly expressed in antler blood slices.In addition,the relative content of 5 triglycerides in three-branch antler wax slices was significantly higher than that in two-branch antler wax slices.In this study,the lipid components of wax and blood slices for two-branched and three-branched antler were analyzed in detail,which will provide theoretical foundation for the exploration of the effective active ingredients of antler and the growth mechanism of antler.
