DRG中P2X3受体参与艾灸足三里穴缓解肠易激综合征大鼠内脏痛的研究 被引量:10

Research of P2X3 Receptor from DRG Participating in Moxibustion at Zusanli(ST36)to Relieve Visceral Pain in IBS Rats

摘要 目的探讨结肠相关背根神经节(dorsal root ganglia, DRG)中P2X3受体参与艾灸足三里穴缓解肠易激综合征(irritable bowel syndrome, IBS)大鼠内脏痛的效应与作用机制。方法清洁级雄性SD新生大鼠随机分为正常组和造模组,采用直结肠球囊扩张(colorectal dilatation, CRD)的方法诱导IBS慢性内脏痛模型。模型鉴定成功后再将造模组大鼠随机分为模型组、艾灸组、P2X3受体拮抗剂组、生理盐水组,艾灸组取双侧足三里穴进行艾灸干预,P2X3受体拮抗剂组进行鞘内注射P2X3受体拮抗剂干预,生理盐水组进行鞘内注射生理盐水干预。运用行为学、免疫组织化学、Western blot、RT-qPCR等方法观察结肠相关背根神经节中P2X3受体在艾灸缓解IBS大鼠内脏痛中的作用。结果与正常组比较,模型组大鼠在20、40、60、80 mmHg不同压力强度CRD刺激下内脏痛行为学评分明显升高,结肠相关DRG中P2X3受体蛋白及其mRNA表达明显增加(均P<0.05);与模型组比较,艾灸组大鼠在20、40、60、80 mmHg不同压力强度CRD刺激下内脏痛行为学评分明显降低,结肠相关DRG中P2X3受体蛋白及其mRNA表达明显减少(均P<0.05)。结论艾灸能够调节IBS大鼠结肠相关DRG中异常增高的P2X3受体蛋白及mRNA表达,可能是艾灸缓解IBS大鼠内脏痛外周敏化的作用机制。 Objective To discuss the effect of P2X3 from colon-associated dorsal root ganglia(DRG)participating in moxibustion at Zusanli(ST36)to relieve visceral pain in irritable bowel syndrome(IBS)rats.Method Clean-grade neonatal male Sprague-Dawley(SD)rats were randomized into a normal group and a model group.IBS chronic visceral pain model was established using neonatal rat colorectal distending(CRD)method.When the model was confirmed a success,the model group rats were randomized into a model group,a moxibustion group,a P2X3 receptor antagonist group and a normal saline group.Bilateral Zusanli points were selected to receive moxibustion intervention in the moxibustion group;the P2X3 receptor antagonist group received intrathecal injection of P2X3 receptor antagonist;the normal saline group received intrathecal injection of normal saline.Behavioral study,and immunohistochemistry,Western Blot(WB),and real-time quantitative polymerase chain reaction(RT-qPCR)methods were adopted to observe the effect of P2X3 receptor from colon-associated DRG in moxibustion treatment to relieve visceral pain in IBS rats.Result Moxibustion at Zusanli significantly reduced the visceral pain-related behavioral score under CRD pressure of 20,40,60 and 80 mmHg in IBS rats.Moxibustion at Zusanli down-regulated the protein and mRNA expressions of P2X3 from colon-associated DRG in IBS rats.Conclusion Moxibustion can down-regulate the abnormally increased P2X3 receptor protein and mRNA expressions from colon-associated DRG in IBS rats,which may be the mechanism of moxibustion in alleviating the peripheral sensitization of visceral pain in IBS rats.
