摘要
目的评价糖尿病大鼠心肌缺血再灌注损伤与线粒体Lon蛋白酶水平的关系。方法健康雄性SD大鼠40只,采用随机数字表法分为4组(n=10):假手术组(sham组)、心肌缺血再灌注组(I/R组)、糖尿病+假手术组(DM+sham组)和糖尿病+心肌缺血再灌注组(DM+I/R组)。DM+sham组和DM+I/R组腹腔注射链脲佐菌素制备糖尿病模型。I/R组和DM+I/R组结扎LAD 30 min再灌注120 min制备大鼠心肌缺血再灌注损伤模型。测定血清cTnI含量、心肌梗死范围、心肌LonP1水平与线粒体顺乌头酸酶(mitochondrial aconitase,mt-ACO2)活性,HE染色观察病理学结果。结果与sham组比较,I/R组和DM+I/R组LonP1水平升高(P<0.05),心肌损伤程度加重(P<0.05),mt-ACO2活性降低(P<0.05),而DM+sham组仅有LonP1水平升高(P<0.05),心肌损伤程度与mt-ACO2活性差异无统计学意义(P>0.05)。与I/R组比较,DM+I/R组心肌损伤程度加重(P<0.05),LonP1水平与mt-ACO2活性降低(P<0.05)。与DM+sham组比较,DM+I/R组心肌损伤程度加重(P<0.05),mt-ACO2活性降低(P<0.05),而LonP1水平差异无统计学意义(P>0.05)。结论糖尿病大鼠心肌缺血再灌注损伤加重可能与糖尿病背景下缺血再灌注损伤针对LonP1的急性诱导能力丧失有关。
Objective To evaluate the relationship between myocardial ischemia-reperfusion injury and mitochondrial Lon protease level in diabetic rats.Methods Forty healthy male SD rats were randomly divided into 4 groups(n=10):sham operation group(sham group),myocardial ischemia-reperfusion group(I/R group),diabetic+sham operation group(DM+sham group)and diabetic+myocardial ischemia-reperfusion group(DM+I/R group).Diabetes model was prepared by intraperitoneal streptozotocin injection in DM+sham group and DM+I/R group.The rat model of myocardial ischemia-reperfusion injury was prepared by ligating LAD for 30 min and reperfusion for 120 min in I/R group and DM+I/R group.Serum cTnI content,myocardial infarction range,myocardial LonP1 level and mitochondrial aconitase(mt-ACO2)activity were determined,and the pathological results were observed with HE staining.Results Compared with sham group,LonP1 level was increased(P<0.05),the degree of myocardial injury was aggravated(P<0.05),and the mt-ACO2 activity was decreased in I/R group and DM+I/R group(P<0.05),while only LonP1 level was increased in DM+sham group(P<0.05),and the degree of myocardial injury and mt-ACO2 activity showed no statistically significant difference(P>0.05).Compared with I/R group,the degree of myocardial injury was aggravated in DM+I/R group(P<0.05),and LonP1 level and mt-ACO2 activity were decreased(P<0.05).Compared with DM+sham group,the degree of myocardial injury was aggravated and the activity of mt-ACO2 was decreased in DM+I/R group(P<0.05),while the LonP1 level showed no statistically significant difference(P>0.05).Conclusion The aggravation of myocardial ischemia-reperfusion injury in diabetic rats may be related to the loss of acute induction of LonP1 after ischemia-reperfusion injury in diabetic rats.
