摘要
目的寻找更优的小鼠骨缺损模型的制作方法,同时动态监测SDF-1在骨缺损部位的表达。为研究SDF-1在骨缺损修复中的作用奠定基础。方法使用打磨机,于小鼠股骨中段制造范围约2 mm×2 mm骨缺损,免疫组化法动态检测小鼠骨缺损部位SDF-1表达情况。结果动物生存情况,除一只在术中死亡外,其余动物均术后存活,进食活动正常,未出现伤口感染情况。骨缺损模型建立后,骨缺损侧SDF-1表达较健侧明显持续增高。骨缺损侧和未手术侧SDF-1表达水平,差异有统计学意义(P<0.05)。结论使用打磨机打磨方法制造骨缺损,过程快速,大小范围较传统骨钳安全有效,可重复性好,动物模型一致性好,可控制性强,为下一步组织工程骨植入提供了较好的动物模型。骨缺损发生后,局部SDF-1分泌增加。在骨缺损修复过程中,SDF-1呈现持续的高表达,说明SDF-1在骨缺损修复过程中的干细胞趋化作用中可能发挥重要作用。
Objective To establish the bone defect model on mice,and detect the expression of SDF-1 during different time phase,in order to understand the process of dynamic changes of SDF-1 expression after bone defect.Me thods We used sander(SAEYANG MARATHON-3)to make 2 mm×2 mm bone defect in the middle of the femur,and detected the expression of SDF-1 during different time phase by immunohistochemistry.Re-s ults sion of SDF-1 was higher on the side of bone defected than the normal side(P<0.05).Conclus ions Using sander to make thigh-bone defect is safe and effective.The expression of SDF-1 is higher on the side of bone defected than normal side.
