意大利牛舌草的化学成分及对大鼠心肌细胞缺氧/复氧损伤的保护作用 被引量:5

摘要 目的研究意大利牛舌草的化学成分及其对大鼠心肌细胞缺氧/复氧损伤的保护作用。方法用体积分数为60%的乙醇提取,经硅胶柱色谱、聚酰胺层析和制备液相等多种色谱方法进行分离纯化得到单体化合物,通过理化性质和核磁共振波谱鉴定化合物结构。建立大鼠乳鼠心肌细胞缺氧/复氧损伤模型,考察单体化合物对心肌细胞存活率、乳酸脱氢酶(LDH)水平、细胞内丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性的影响。结果从意大利牛舌草中分离鉴定了10种化合物,分别为β-谷甾醇乙酸酯(1)、β-谷甾醇(2)、苦莓苷(3)、芦丁(4)、山柰酚(5)、咖啡酸(6)、胡萝卜苷(7)、山柰酚-3-O-芸香糖苷(8)、水仙苷(9)和迷迭香酸(10)。化合物4,8,9和10组心肌细胞存活率分别为91.05%,92.32%,93.41%和93.38%。与模型组比较,心肌细胞LDH水平和MDA含量均显著降低,SOD活性显著升高。结论化合物1,2,3,7和8均为首次从该植物中分离得到,单体化合物4,8,9和10对缺氧/复氧心肌细胞损伤具有保护作用。 Objective To study the chemical constituents of Anchusa italica Retz.and protective effects on hypoxia/reoxygenation induced oxidative stress injury in rat primary cardiomyocyte.Methods Chemical compositions of Anchusa italica Retz.were extracted with 60%ethanol,and were separated and purified by various chromatographic methods such as silica gel column chromatography,polyamide column chromatography and preparative liquid chromatography,respectively,and the structures of the purified compounds were identified by physicochemical properties analysis and NMR.Rat primary cardiomyocytes were cultured in vitro and hypoxia/reoxygenation induced oxidative stress injury model was established.The effects of selected compounds on myocardial cell viability,lactate dehydrogenase(LDH)level,intracellular malondialdehyde(MDA)content and intracellular superoxide dismutase(SOD)activity were investigated.Results 10 compounds were isolated from Anchusa italica Retz.,and the structures of compounds were identified asβ-sitosterol acetate(1),β-sitosterol(2),nigaichigoside(3),rutin(4),kaempferol(5),caffeic acid(6),daucosterol(7),kaempferol-3-O-rutinoside(8),narcissin(9)and rosmarinic acid(10).Cell survival rates of compounds 4,8,9 and 10 groups were 91.05%,92.32%,93.41%and 93.38%,respectively.Compared with the model group,the contents of LDH and MDA decreased significantly,and the activity of SOD increased significantly.Conclusion Compounds 1,2,3,7 and 8 were obtained from Anchusa italic Retz.for the first time.Compounds 4,8,9 and 10 have protective effects on hypoxia/reoxygenation induced oxidative stress injury in rat primary cardiomyocyte.
