摘要 PD⁃1/PD⁃L1在肿瘤免疫逃逸中的作用机制是最近研究热点,靶向PD⁃1/PD⁃L1途径被认为是癌症治疗的有力的策略。然而,但这些药物的疗效和反应性在不同的肿瘤类型和不同的患者中差异很大。这引起人们对PD⁃L1表达调控机制的研究兴趣。PD⁃L1在肿瘤细胞中的表达机制是多层次的,包括基因组扩增、组成性癌基因信号激活、外源性因素和表观遗传机制。越来越多研究表明,PD⁃L1高表达可能是影响抗PD⁃1/PD⁃L1治疗效果的重要因素之一,并与更高的癌症死亡风险和不良的患者预后相关。深入了解PD⁃L1在肿瘤细胞中表达的调节机制,为今后肿瘤免疫治疗提供新的可能。 The mechanism of PD⁃1/PD⁃L1 in tumor immune escape has been a hot research topic recently.Targeting the PD⁃1/PD⁃L1 pathway is considered as a powerful strategy for cancer therapy,but the efficacy and responsiveness of these drugs vary widely among different tumor types and patients.This leads to research interest in the regulation of PD⁃L1 expression.The mechanism of PD⁃L1 expression in tumor cells is multi⁃layered,including genomic alterations(amplification and regulation),signal activation of constitutive oncogenes,exogenous factors and epigenetic mechanisms.A growing body of research suggests that high expression of PD⁃L1 may be an impor⁃tant factor in the efficacy of anti⁃PD⁃1/PD⁃L1 therapy and is associated with more aggressive and malignant tumor subtypes,higher cancer mortality risk,and poor patient outcomes.Our research can help understand the regulatory mechanism of PD⁃L1 expression in tumor cells and provide new possibilities for tumor immunotherapy in the future.
