摘要
核酸序列依赖性扩增技术(nucleic acid sequence-based amplification,NASBA)是以PCR为基础发展的恒温扩增技术,操作简单、高保守性、高灵敏度、高特异性、引物设计简单、检测周期短和兼容性强,适用于RNA的扩增,能在2 h将单链RNA扩增约109倍,无需特殊设备。NASBA技术已广泛应用于呼吸道病毒、肠道病毒、食物传播病毒、免疫缺陷病毒和动物源性病毒等病原体的检测。本文就NASBA技术的检测原理、技术特点、扩增产物检测方法以及在蚊媒传染病检测中的应用做一总结。
Nucleic acid sequence-based amplification(NASBA)is a constant temperature amplification technology developed on the basis of PCR.It has the advantages of simple operation,high conservation,high sensitivity,and specificity,simple primer design,short detection cycle and strong compatibility.It is suitable for RNA amplification.It can amplify template single strand RNA about 109 times in 2 hours without special equipment.NASBA technology has been widely used in the detection of respiratory virus,enterovirus,food-borne virus,immunodeficiency virus and animal derived virus.In this paper,the detection principle,technical characteristics,detection of amplification products and the application of NASBA in the detection of mosquito-borne infectious diseases had been summarized.
