刃针疗法对膝骨关节炎白兔膝关节软骨组织黏着斑激酶/丝裂原活化蛋白激酶信号通路的调控作用 被引量:9

摘要 目的探讨刃针疗法对膝骨关节炎(KOA)白兔膝关节软骨组织黏着斑激酶(FAK)/丝裂原活化蛋白激酶(MAPK)信号通路的调控作用。方法选取54只白兔,采用前交叉韧带切断术建立KOA模型;将造模成功的48只白兔随机分为模型组、刃针组、电针组、模型抑制组、刃针抑制组、电针抑制组,每组8只,另纳入8只白兔作为空白组。空白组和模型组不予治疗;刃针组和刃针抑制组均采用刃针治疗,电针组和电针抑制组均采用电针治疗;刃针抑制组和电针抑制组均在治疗前2 h给予关节腔注射抑制剂PF-562271,模型抑制组同时给予关节腔注射抑制剂。干预结束后,取各组白兔膝关节软骨组织,检测整合素β1、FAK、细胞外调节蛋白激酶(ERK)1/2、c-Jun氨基末端激酶(JNK)和p38的蛋白表达水平。结果与空白组对比,模型组整合素β1、FAK、p38、JNK及ERK1/2蛋白表达水平升高(均P<0.05)。与模型组比较,刃针组、电针组整合素β1、FAK、p38、JNK及ERK1/2蛋白表达水平均降低(均P<0.05)。与电针组对比,刃针组整合素β1、JNK及ERK1/2蛋白表达水平降低(均P<0.05),而两组FAK、p38表达水平比较均无明显差异(均P>0.05)。与相应干预方法组对比,使用抑制剂的各组整合素β1、FAK、p38、JNK及ERK1/2蛋白表达水平均下降(均P<0.05)。结论刃针能够抑制过度活跃的FAK-MAPK信号通路,这或是其促进关节软骨修复的可能力学机制。 Objective To investigate the regulatory effect of blade needle therapy on focal adhesion kinase(FAK)/mitogen-activated protein kinase(MAPK)signaling pathway in knee cartilage tissues of rabbits with knee osteoarthritis(KOA).Methods Fifty-four rabbits were selected for establishing KOA models by anterior cruciate ligament transection;then 48 rabbits with successful modeling were randomized to model group,blade needle group,electropuncture group,model inhibition group,blade needle inhibition group or electropuncture inhibition group,with 8 rabbits in each group,and another 8 rabbits were selected as blank group.No treatment was performed in the blank group and the model group;blade needle therapy was conducted in the blade needle group and the blade needle inhibition group,while electropuncture therapy in the electropuncture group and the electropuncture inhibition group;the blade needle inhibition group and the electropuncture inhibition group were treated by intra-articular injection of inhibitor PF-562271 two hours before treatment,and intra-articular injection of inhibitor was also conducted in the model inhibition group concurrently.After intervention,the knee cartilage tissues were removed from the rabbits of all groups,then the protein expression levels of integrinβ1,FAK,extracellular regulated protein kinase(ERK)1/2,c-Jun N-terminal kinase(JNK)and p38 were determined.Results Compared with the blank group,the model group had elevated expression levels of integrinβ1,FAK,p38,JNK and ERK1/2 proteins(all P<0.05).Compared with the model group,the blade needle group and the electropuncture group yielded lower expression levels of integrinβ1,FAK,p38,JNK and ERK1/2 proteins(all P<0.05).The blade needle group exhibited lower expression levels of integrinβ1,JNK and ERK1/2 proteins as compared with the electropuncture group,furthermore,there was no significant difference between the two groups in expression level of FAK or p38(all P<0.05).The groups using inhibitors obtained lower expression levels of integrinβ1,F
