摘要
为了研究杨梅黄酮对环磷酰胺所致雄性小鼠生精障碍的保护作用及作用机制,采用腹腔注射环磷酰胺(50 mg/kg·bw)连续7 d建立雄性小鼠生精障碍模型,杨梅黄酮组分别灌胃杨梅黄酮(100 mg/kg·bw、200 mg/kg·bw、400 mg/kg·bw),连续30 d。观察雄性小鼠的精子密度、精子畸变率,血清中还原型谷胱甘肽(GSH)的含量,睾丸组织中乳酸脱氢酶(LDH)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)及核因子κB(NF-κB)等指标,并观察睾丸组织的病理改变。结果显示:与模型组比较,杨梅黄酮中、高剂量组的精子密度增加、精子畸变率减少,杨梅黄酮中、高剂量组睾丸组织中TNF-α及NF-κB含量降低,杨梅黄酮中、高剂量组血清中GSH水平增加,杨梅黄酮低、中、高剂量组睾丸组织中MDA含量降低、LDH活性增强。病理可见模型组睾丸组织生精上皮明显变薄,生精细胞层次、数量减少,多数生精小管腔未见精子形成,而杨梅黄酮可改善环磷酰胺所致的生精细胞的损伤。表明杨梅黄酮对环磷酰胺诱导雄性小鼠的生精障碍有一定保护作用,作用机制与抗氧化、抗炎有关。
In this study,we investigated the protective effect of myricetin on the reproductive function and its mechanism for reproductive injury in male mice caused by cyclophosphamide.The reproductive toxicity of male mice model was built by cyclophosphamide intraperitoneal injection(50 mg/kg·bw)for seven days.The mice were intragastrically treated with myricetin solutions at 100 mg/kg·bw,200 mg/kg·bw and 400 mg/kg·bw for the treatment groups once a day for 30 consecutive days,respectively.The mice were sacrificed on day 31 for the measurement of the sperm density,the sperm aberration rate.The content of reduced glutathione(GSH)in blood serum,the activity of lactate dehydrogenase(LDH),the contents of malondialdehyde(MDA),tumor necrosis factor-α(TNF-α)and nuclear factor kB(NF-κB)in testicular tissue homogenate were assayed respectively.The pathological changes of testicular tissues were observed.Compared with the model group.The sperm density and the content of GSH in blood serum were increased,the sperm aberration rate mice,and the contents of TNF-α and NF-κB in testicular tissue homogenate were decreased in 200 mg/kg·bw,and 400 mg/kg·bw myricetin groups.Theactivity of LDH in testicular tissue homogenate was increased and the content of MDA in testicular tissue homogenate was decreased in different dose myricetin groups.The construction of testicular tissues was damaged,and sperms and interstitial cells of testicles decreased significantly.Cell number of spermatogenic epithelium in model group were lower than those in control group on morphological testis.There was decline of spermatogenic cells in levels and numbers and no spermiogenesis in most of seminiferous tubule in model group.The change above could be reversed by Myricetin.Myricetin has certain protective effects on spermatogenesis dysfunction of mice caused by cyclophosphamide,and its mechanism was related to anti-oxidation and anti-inflammation.
