摘要
为了解山东地区2017-2018年猪流行性腹泻病毒(PEDV)的流行情况,通过RT-PCR方法对山东省各地疑似PEDV病料进行检测,并对阳性样本进行ORF3基因的扩增和遗传进化分析。结果显示,28份具有代表性的PEDV ORF3序列,大小为675 bp,编码224个氨基酸。遗传进化分析显示,28株PEDV ORF3不存在碱基增加或缺失,出现一定规律的碱基突变。28株ORF3基因同源性为94.2%~100%,编码氨基酸同源性为94.7%~99.6%。遗传分析表明,2017-2018年山东省流行的PEDV毒株分为2个亚群,其中22株流行株位于2-1亚群,与韩国病毒株存在一定遗传变异关系,其余6株与南方流行毒株聚类在一起。该试验为持续追踪PEDV的变异趋势和开发PEDV新型疫苗奠定理论基础。
In order to understand the prevalence of porcine epidemic diarrhea virus(PEDV)in Shandong from 2017 to 2018,the suspected PEDV disease was detected by RT-PCR method,and the positive samples were confirmed by amplifying ORF3 gene.Its genetic evolution was also analyzed.The results showed that the size of 28 representative PEDV ORF3 sequences was 675 bp,encoding 224 amino acids.Genetic evolution analysis showed that 28 strains of PEDV ORF3 did not have base deletions,but only mutations of individual bases.The homology of 28 strains of ORF3 gene was 94.2%~100%,and the amino acid homology was 94.7%~99.6%.Genetic analysis showed that PEDV strains were divided into two groups.The prevalence of PEDV in Shandong Province was mainly located in the second group,which is closely related to Korean virus strains,and 6 strains were closely related to the strain in South China.This study lays the foundation for further study of PEDV heritable variation and development of new PEDV vaccines and diagnostic reagents.
