摘要
目的 探究益气温阳方对慢性心力衰竭(CHF)大鼠心功能改善的作用及其机制。方法 选取75只SD大鼠,采用随机数字表法将大鼠分为健康对照组、模型组、低剂量益气温阳方组、中剂量益气温阳方组、高剂量益气温阳方组,每组各15只。除健康对照组外各组采用肾上腹主动脉缩窄法建立CHF模型。低剂量益气温阳方组大鼠灌胃剂量为6. 9 g/(kg·d)、中剂量益气温阳方组大鼠灌胃剂量为13. 7 g/(kg·d)、高剂量益气温阳方组大鼠灌胃剂量为27. 4g/(kg·d),每日一次,共灌胃8周;健康对照组和模型组使用27. 4 g/(kg·d)的生理盐水灌胃处理;检测各组大鼠左心室收缩压(LVSP)、左心室舒张压(LVEDP)、左心室内压上升下降最大速率(±LVdp/dtmax);采用HE染色观察大鼠心肌细胞状态;采用高压液相色谱法检测各组大鼠心肌细胞中三磷酸腺苷(ATP)、二磷酸腺苷(ADP)、单磷酸核糖核苷酸(AMP)含量;采用ELISA法白细胞介素-6(IL-6)、诱生型一氧化氮合酶(i NOS)的含量;采用蛋白质印记法检测各组大鼠心肌细胞凋亡蛋白Caspase-3和Caspase-9蛋白表达水平。结果 HE染色实验可以看出,模型组心肌肌纤维弯曲、肌红蛋白溶解,肌细胞核固缩、溶解;相比模型组,使用益气温阳方处理后各组大鼠心肌外膜较为完整、肌纤维断裂溶解显著改善。相比健康对照组,模型组大鼠LVSP、+LVdp/dtmax、-LVdp/dtmax、IL-6、i NOS、Caspase-3、Caspase-9表达显著升高(P <0. 05),LVEDP、ATP、ADP、AMP含量水平显著降低(P <0. 05);相比模型组,使用益气温阳方处理各组大鼠LVSP、+LVdp/dtmax、-LVdp/dtmax、IL-6、i NOS、Caspase-3、Caspase-9表达显著降低(P <0. 05),且在本实验浓度范围内呈剂量依赖性,LVEDP、ATP、ADP、AMP含量水平显著升高(P <0. 05),且在本实验浓度范围内呈剂量依赖性。结论 益气温阳方可以改善CHF大鼠心脏血流动力学,同时优化能量代谢,抑制心肌炎症反应,保护心
Objective To explore improvement effect of Yiqi Wenyang prescription(YWP)on cardiac function of chronic heart failure(CHF)rats and its mechanism.Methods Seventy-five SD rats were enrolled.They were divided into healthy control group,model group,low-dose,medium-dose and high-dose YWP group by random number table method,15 cases in each group.Except healthy control group,CHF models were established by adrenal abdominal aorta coarctation in the other groups.The left ventricular systolic pressure(LVSP),left ventricular end-diastolic pressure(LVEDP)and the maximum rate change of left ventricular pressure(±LVdp/dtmax)in each group were detected.The cardiomyocytes status was observed by HE staining.The high pressure liquid chromatography was applied to detect contents of ATP,ADP and AMP in cardiomyocytes.ELISA was applied to detect contents of interleukin-6(IL-6)and inducible nitric oxide synthase(iNOS).Western blot was applied to detect expression levels of cardiomyocytes apoptin Caspase-3 and Caspase-9 proteins in each group.Results HE staining experiment showed that myocardial muscle fibers were crooked,myoglobin was dissolved,and muscle cytoblast was condensed and dissolved in model group.Compared with model group,myocardial adventitia was more complete,breaking and dissolving of muscle fibers were more significant in all groups treated with Yiqi Wenyang prescription.Compared with healthy control group,LVSP,+LVdp/dtmax,-LVdp/dtmax,IL-6,iNOS,expression of Caspase-3 and Caspase-9 were significantly increased in model group(P<0.05),while contents of LVEDP,ATP,ADP and AMP were significantly decreased(P<0.05).Compared with model group,LVSP,+LVdp/dtmax,-LVdp/dtmax,IL-6,iNOS,expression of Caspase-3 and Caspase-9 were significantly decreased in all groups treated with Yiqi Wenyang prescription,showing dose-dependence within the concentration range(P<0.05),while contents of LVEDP,ATP,ADP and AMP were significantly increased,showing dose-dependence within the concentration range(P<0.05).Conclusion Yiqi Wenyang prescription c
