摘要
目的探讨血小板贮存期间血小板扩散功能和炎症介质的变化。方法选择泰州市中心血站单采血小板,分别于第1、3、5天进行扩散试验,并检测血栓素B2(TXB2)、前列腺素E2(PGE2)、血小板因子4(PF4)、可溶性CD40L(SCD40L)、CD62P和活性氧(ROS)等炎症介质变化。结果与第1天比较,第3、5天血小板的扩散能力下降[(25±7)%、(15±3)%vs.(52±12)%](P<0.01),第5天血小板的扩散能力低于第3天(P<0.01)。与第1天比较,第3、5天的TXB2[(1480±282)pg/mL、(3570±650)pg/mL vs.(605±58)pg/mL]、PGE2[(11000±2480)pg/mL、(10658±1874)pg/mL vs.(4930±1270)pg/mL]、PF4[(3558±962)ng/mL、(4010±1027)ng/mL vs.(1226±205)ng/mL]、SCD40L[(1106±239)pg/mL、(1248±383)pg/mL vs.(482±55)pg/mL]、CD62P[(62±9)%、(95±2)%vs.(51±5)%]和ROS(150±31、211±39 vs.80±6)均增加(P<0.01),第5天的TXB2、CD62P和ROS均高于第3天(P<0.01)。结论在贮存期间,单采血小板扩散功能逐日下降,炎症介质不断升高,可能会影响临床治疗效果。
Objective To investigate the changes of diffusion function and inflammatory mediators of platelets during storage.Methods The single blood platelets samples were collected from Taizhou City Center Blood Station.The diffusion function of platelets was tested on the 1st,3rd and 5th day of storage and the inflammatory mediators of platelets were detected at the same times,which included thromboxane B2(TXB2),prostaglandin E2(PGE2),platelet factor 4(PF4),soluble CD 40 L(SCD40 L),CD62 P and active oxygen(ROS).Results Compared to the 1st day of storage,the diffusion function of platelets was significantly decreased on the 3rd and 5th day of storage[(25±7)%and(15±3)%vs.(52±12)%](P<0.01),which was lower on the 5th day than that on the 3rd day of storage(P<0.01).Compared to the 1st day of storage,the levels of TXB2[(1480±282)pg/mL and(3570±650)pg/mL vs.(605±58)pg/mL],PGE2[(11000±2480)pg/mL and(10658±1874)pg/mL vs.(4930±1270)pg/mL],PF4[(3558±962)ng/mL and(4010±1027)ng/mL vs.(1226±205)ng/mL],SCD40 L[(1106±239)pg/mL and(1248±383)pg/mL vs.(482±55)pg/mL],CD62 P[(62±9)%and(95±2)%vs.(51±5)%]and ROS(150±31 and 211±39 vs.80±6)were all significantly increased(P<0.01),which of TXB2,CD62 P and ROS on the 5th day were higher than those on the 3rd day of storage(P<0.01).Conclusion During the storage period,the diffusion function of single blood platelets is decreased day by day,and the inflammatory mediators are increased continuously,which may affect the clinical treatment effect.
