青海贵南某牧场犊牦牛腹泻病原的分子鉴定分析 被引量:7

Molecular Identification and Analysis Pathogens Causing Young Yak Diarrhea in a Pasture in Guinan of Qinghai Province

摘要 青海省贵南县某牧场发生犊牦牛腹泻疾病,为快速确诊发病犊牦牛腹泻的致病原因,及时防控和治疗,采集犊牦牛腹泻样品15份,利用形态学方法和分子生物学方法进行鉴定与分析。结果显示:引发该牧场犊牦牛腹泻的病原是隐孢子虫(Cryptosporidium)和邱氏艾美耳球虫(Eimeria zurnii,E.zurnii)、牛艾美耳球虫(Eimeria bovi,E.bovi)、椭圆艾美耳球虫(Eimeria ellipsoidalis,E.ellipsoidalis)的混合感染,测序结果显示,瑞氏隐孢子虫(Cryptosporidium ryanae,C.ryanae)的18S rRNA基因与参考序列中国株CP031554/3/2/1同源性在99%以上,各个种球虫与引用参考序列的同源性均在98%以上。同时,遗传进化树显示:本次检测的C.ryanae与中国株C.ryanae 18S rRNA基因同源性关系近,聚成一大支;三种球虫分别与其相同种聚集在一支上。结果表明,该牧场犊牛腹泻是由隐孢子虫和球虫的混合感染造成的,该研究为犊牛腹泻的病原学研究和流行病学调查提供了参考数据,为寄生虫性病原的对症治疗、对因治疗和辅助支持治疗提供了帮助。 There was an yak diarrhea disease occurring in a pasture in Guinan County of Qinghai Province.For the rapid diagnosis of the causes of yak diarrhea,and timely prevention and treatment of the diarrhea,this study was carried out.Fifteen yak diarrhea samples were collected,the morphological method and molecular method were used for identification and analysis.The results showed that the pathogens causing diarrhea in this pasture were Cryptosporidium and Coccidia(Eimeria zurnii,E.bovi and E.ellipsoidalis).The sequencing results showed that the same pathogens were identified in the samples,the homology of Cryptosporidium ryanae and the reference sequences Chinese strain CP031554/3/2/1 was more than 99%based on the 18 S rRNA gene,the homology of each species of coccidia with the reference sequences was more than 98%respectively.While the genetic trees showed that C.ryanae clustered into a large branch with high homology,the three coccidia were respectively clustered with the same species.The yak diarrhea in Guinan pasture was caused by a mixed infection of Cryptosporidium and coccidian.This study provided reference data for the pathogenic research and epidemiological investigation of yak diarrhea,and also provided helpful reminder for symptomatic treatment,etiological treatment and auxiliary support to the parasitic pathogens.
